Since the capacity for photo protection is limited, drought can lead to damaged and loss of active PSII reaction centers. Therefore, non-destructive measurements of photosynthesis by chlorophyll a fluorometry may potentially provide to plant viability and performance in response to drought. Rice sensitive to drought increase when drought happen during the vegetative and flowering period. plants response to drought stress by identification of character that an important role in drought tolerance can be assessment in levels of morphological, cellular, biochemical and molecular. This experiment was conducted to evaluate the rice genotypes by measuring the traits associated with photosynthetic pigments and chlorophyll fluorescence components in order to select appropriate criteria for screening and drought stress tolerant genotypesMaterials and Methods: This experiment was conducted completely randomized design in two condition of well watered and drought stress during seedling stage in rice genotypes with 56 different provinces of Iran and the International Rice Research Institute (IRRI) in Iran Rice Research Institute (Amol) in 2014. After seeding, irrigation was carried out on a daily basis up to two to three leaf seedling stage. After 15 days, in Customary irrigated conditions, irrigation continued to end of the growth period and was completely cut off under irrigation stress. After soil moisture reached 20% (about 10 days after irrigation stop), the traits of photosynthetic pigments such as chlorophyll a, b and carotenoids and chlorophyll fluorescence components such as Fv / Fm, ΦPSII, q N, ETR and PAR was measured on the basis of scientific methodsResult and Discussion:The results showed that the amount of chlorophyll a and b, respectively, 2.3 and 21.4 percent increase and carotenoids content, 3.8 percent decline in drought stress. Chlorophyll fluorescence parameters except Fv: Fm showed significant differences in the two environments. ΦPSII, qN, ETR and PAR parameters, respectively, the value of 7.3, 13, 35.5 and 28.8 percent were reduced in stress to normal condition. Correlation results showed that chlorophyll a with b and carotenoids with chlorophyll a and b had significant positive correlation. Correlation between ΦPSII with Fv:Fm and qN to correlation coefficient 0.29 and - 0.91 was significant, respectively, . Conclusions:The results showed that under drought stress can be using chlorophyll pigments and chlorophyll fluorescence parameters, especially ΦPSII and ETR as evaluation criteria for the selection and screening of tolerant rice genotypes utilized}, keywords = {Rice,maximum quantum yield,Chlorophyll fluorescence,photosystem II,Screening}, title_fa = {ارزیابی و انتخاب ژنوتیپ‌های متحمل به خشکی برنج با کاربرد روش‌های فلئورومتری}, abstract_fa = {سابقه و هدف: کمبود آب خاک و اتمسفر مهمترین فاکتور محدودکننده فتوسنتز و رشد گیاه به ویژه برنج بوده و بر فتوسنتز از طریق کاهش جذب کربن به واسطه محدودیت متابولیکی و روزنه ای اثرات منفی ایجاد می کند.. با توجه به محدود ظرفیت کربن گیری گیاه، خشکی منجر به خسارت یا تلفات مراکز واکنش فعال در فتوسیستم دو می گردد. بنابراین اندازه گیری غیر تخریبی فتوسنتز ازطریق فلئورومتری کلروفیلa، پتانسیل زنده ماندن گیاه و علائمی از پاسخ به خشکی را فراهم می نماید. برنج به افزایش خشکی وقتی که در دوره رویشی و گلدهی اتفاق می افتد حساس است. پاسخ گیاهان به تنش خشکی از طریق شناسایی خصوصیاتی که نقش مهمی در تحمل به تنش خشکی در سطوح مورفولوژیکی، فیزیولوژیکی، سلولی، بیوشیمیایی و مولکولی بازی می‌کنند ارزیابی می‌گردد. این آزمایش با هدف ارزیابی ژنوتیپ های برنج با ‌اندازه‌گیری صفات مرتبط با ‌رنگزه‌های فتوسنتزی و مولفه‌های فلئورسانس کلروفیل به منظور انتخاب معیار مناسب برای غربالگری و ژنوتیپ‌های متحمل به تنش خشکی انجام شده است. مواد و روش‌ها: آزمایش به صورت طرح کاملاً تصادفی دردو شرایط آبیاری کامل و تنش طی مرحله گیاهچه‌ای با 56 ژنوتیپ برنج ازاستان های مختلف ایران و موسسه تحقیقات بین المللی برنج (IRRI) در سال 1393 در موسسه تحقیقات برنج کشور (آمل) اجرا شد. بعد از بذرپاشی، آبیاری به صورت روزانه تا مرحله دو تا سه برگی گیاهچه انجام شد. بعد از15روز، درشرایط بدون تنش، آبیاری تا پایان دوره رشد ادامه و در محیط تحت تنش آبیاری به طور کامل قطع شد . پس ازاینکه رطوبت خاک به 20 درصد رسید (حدود 10 روز بعد از قطع آبیاری)، صفات مربوط به رنگیزه‌های کلروفیل مانند کلروفیل a، b و کارتنوئیدها و همچنین مولفه‌های فلئورسانس کلروفیل مانند Fv/ Fm، ΦPSII ، qN، ETR به همراه PAR بر اساس روش‌های علمی اندازه‌گیری شد. یافته‌ها: نتایج نشان داد که مقادیرکلروفیل a و b به ترتیب به مقدار 3/2 و 4/21 درصد در تنش خشکی افزایش و مقدارکارتنوئیدها 8/3 درصد کاهش داشت. مولفه‌های فلئورسانس کلروفیل به جز Fv/ Fm اختلاف‌آماری معنی‌داری در دو محیط نشان دادند. مولفه‌های ΦPSII qN ETR و PAR به ترتیب به مقدار 7/3، 13، 5/35 و 8/28 درصد در شرایط تنش نسبت به نرمال کاهش داشتند. نتایج همبستگی صفات نشان داده که بین کلروفیل a با b و کارتنوئید با کلروفیل a و b همبستگی مثبت و معنی‌دار وجود داشت. همبستگی بین ΦPSII با Fv/ F m و با q N به‌ترتیب با ضریب همبستگی 29/0 و 91/0- معنی‌دار بوده است. نتیجه گیری: نتایج حاصل از این تحقیق حاکی از آن است که از رنگیزه‌های کلروفیل a، b، کارتنوئیدها و مولفه‌های فلئورسانس کلروفیل به‌ویژه ΦPSII و ETR می‌توان به عنوان معیارهای ارزیابی برنج در مرحله گیاهچه جهت انتخاب و غربال‌گری ژنوتیپ‌های متحمل به خشکی استفاده کرد.}, keywords_fa = {برنج,حداکثر عملکرد کوآنتومی,فلئورسانس کلروفیل,فتوسیستم دو,غربال گری}, url = {https://jopp.gau.ac.ir/article_5242.html}, eprint = {https://jopp.gau.ac.ir/article_5242_3436187f1a7aa5886e4c95a18cba4b24.pdf} }
