بررسی عملکرد، شاخصهای رقابت و سودمندی اقتصادی الگوی کاشت مخلوط گیاه دارویی سرخارگل (Echinacea purpurea Moench.) و کنجد (Sesamum indicum L.) در شرایط اقلیمی متفاوت استان مازندران
هدف پژوهش حاضر ارزیابی واکنش گیاه دارویی سرخارگل (Echinacea purpurea Moench.) در کشت مخلوط با کنجد (Sesamum indicum L.) به تغییرات خرداقلیمی استان مازندران بود. آزمایش در قالب طرح بلوکهای کامل تصادفی با سه تکرار در دو منطقه سیمرغ و سوادکوه استان مازندران در سال زراعی 1394 اجرا شد. تیمارها شامل تککشتی سرخارگل، 75% سرخارگل+25% کنجد، 50% سرخارگل+50% کنجد، 25% سرخارگل+75 کنجد و تککشتی کنجد بود. در این پژوهش، برخی شاخصها نظیر نسبت برابری زمین (LER) و سودمندی مالی (MAI) کشت مخلوط و ضریب غالبیت (A) و افت یا افزایش عملکرد واقعی (IYA or LYA) دو گونه گیاهی بر اساس عملکرد دانه کنجد و زیستتوده سرخارگل ارزیابی شد. تجزیه آماری دادههای آزمایش با استفاده از نرمافزار SAS و مقایسه میانگینها برای صفات مورد ارزیابی بهروش آزمون چند دامنهای دانکن در سطح احتمال پنج درصد انجام شد.یافتهها: در این پژوهش، تککشتی سرخارگل در منطقه سوادکوه از حداکثر تولید (3631 کیلوگرم در هکتار) برخوردار بود که در مقایسه با منطقه سیمرغ حدود هفده درصد افزایش نشان داد. همچنین، کشت مخلوط 75% سرخارگل+25% کنجد از عملکرد اقتصادی مشابهی در دو منطقه برخوردار بود و حداقل تولید اقتصادی به تککشتی کنجد در منطقه سوادکوه (1716 کیلوگرم در هکتار) تعلق داشت. با توجه به شاخصهای رقابتی دو گونه، سرخارگل (Apc) و کنجد (Ase) در گسترهی عددی بین 16/0-02/0 Apc=و 26/0-10/0 Ase= بهترتیب در منطقه سوادکوه و سیمرغ گونه غالب بودند. با توجه به افت شدید حداقل دما و رطوبت نسبی اقلیم سوادکوه در مقایسه با منطقه سیمرغ طی فصل رشد، شاخص غالبیت سرخارگل مؤید برخوراری گیاه از بهترین شرایط محیطی و به تبع آن افزایش تجمع ماده خشک در این منطقه میباشد. علاوهبر این، حداکثر شاخص عملکرد واقعی سرخارگل به کشت مخلوط 50% سرخارگل+50% کنجد در منطقه سوادکوه (23/0AYLpc=) تعلق داشت و افت عملکرد واقعی آن در آرایش کاشت 25% سرخارگل+75% کنجد در منطقه سیمرغ (14/0-AYLpc=) مشاهده شد. همچنین، حداکثر افزایش عملکرد واقعی کنجد به کشت مخلوط 25% کنجد+75% سرخارگل در منطقه سیمرغ (26/0AYLse=) تعلق داشت. نتایج بیانگر آن است که ترکیبهای مختلف کشت مخلوط در یک گروه آماری افزایش بیش از هفت درصدی نسبت برابری زمین را در مقایسه با تککشتی گونهها موجب گردیدند و کشت مخلوط 75% سرخارگل+25% کنجد از بالاترین شاخص سودمندی مالی کشت مخلوط (1823MAI=) برخوردار بود. نتیجهگیری: در مجموع، نتایج بیانگر تأثیرپذیری کشت مخلوط از شرایط خرداقلیمی دو منطقه بود. بر این اساس، کنجد و سرخارگل با سهم 25 و 50 در کشت مخلوط بهترتیب در مناطق سیمرغ و سوادکوه از توانایی رقابتی بالاتری برخوردار بودند.

تجزیه عاملی کارآیی مصرف آب و برخی صفات کمی و عملکرد در کنجد (Sesamum indicum L.) تحتتأثیر کاربرد نیتروژن و نهادههای اکولوژیک در شرایط تنش خشکی
چکیدهسابقه و هدف: دسترسی ناکافی به آب در مناطق خشک و نیمهخشک، افزایش کارآیی مصرف آب را بهعنوان یکی از محورهای اصلی کشاورزی پایدار در این مناطق مطرح کرده است. از این رو، در سالهای اخیر تلاشهای فراوانی بهمنظور افزایش کارایی مصرف آب صورت گرفته است که در این مسیر، بهرهگیری از سوپرجاذبها و محلولپاشی نهادههای بومسازگاری نظیر اسید هیومیک و اسید سالیسیلیک جهت صرفهجویی و استفادهی بهینهی آب مدنظر قرار گرفتهاند، لذا این پژوهش با هدف استفاده از تجزیه و تحلیل عاملی در تعیین عوامل مؤثر بر کارآیی مصرف آب کنجد در شرایط کاربرد همزمان نیتروژن و نهادههای بومسازگار انجام شد. مواد و روشها: این پژوهش در سال زراعی 95- 1394 در مزرعه تحقیقاتی دانشگاه فردوسی مشهد بهصورت کرتهای خرد شده در قالب طرح پایهی بلوکهای کامل تصادفی با سه تکرار انجام شد. سطوح آبیاری (50 و 100 درصد نیاز آبی) در کرتهای اصلی و تیمارهای تغذیهای شامل 1- کاربرد 75 کیلوگرم در هکتار نیتروژن خالص بهعلاوه 80 کیلوگرم سوپرجاذب رطوبت، 2- کاربرد 80 کیلوگرم در هکتار هیدروژل سوپرجاذب رطوبت بهعلاوه کاربرد توأم 6 کیلوگرم در هکتار اسید هیومیک و یک میلیمولار اسید سالیسیلیک 3- کاربرد توأم 6 کیلوگرم در هکتار اسید هیومیک و یک میلیمولار اسید سالیسیلیک بهعلاوه کاربرد 75 کیلوگرم در هکتار نیتروژن خالص 4- کاربرد توأم 6 کیلوگرم در هکتار اسید هیومیک و یک میلیمولار اسید سالیسیلیک بهعلاوه 75 کیلوگرم در هکتار نیتروژن خالص بهعلاوه 80 کیلوگرم سوپرجاذب رطوبت و 5- شاهد در کرتهای فرعی قرار گرفتند.یافتهها: نتایج آزمایش نشان داد که بیشترین عملکرد دانه (1672 کیلوگرم در هکتار) با تأمین 100 درصد نیاز آبی و در تیمار کاربرد همزمان نیتروژن، سوپرجاذب، اسید هیومیک و اسید سالیسیلیک بدست آمد. نقش اسید هیومیک و اسید سالیسیلیک در بهبود اکثر صفات مورد مطالعه بسیار محسوس بود، بهطوریکه عملکرد دانه، شاخص سطح برگ، سرعت رشد محصول، و کارآیی مصرف آب بهترتیب 22، 31، 30 و 21 درصد در تیمارهای دارای اسید هیومیک و اسید سالیسیلیک بیشتر از تیمارهای بدون این دو نهاده بود. بر اساس نتایج تحلیل عاملی، متغیرها به دو عامل تجزیه شده و عامل اول 59 درصد از واریانس متغیرها را تبیین کرد. عامل اول شامل متغیرهای عملکرد دانه، وزن دانه در بوته، سرعت رشد محصول، نیتروژن و فسفر خاک و کارآیی مصرف آب و عامل دوم به متغیرهای عملکرد ماده خشک، ارتفاع بوته، شاخص سطح برگ و pH خاک تعلق گرفت. نتیجهگیری: بهطور کلی نتایج این پژوهش نشان داد که در شرایط تنش خشکی (تأمین 50 درصد نیاز آبی)، تیمارهای کاربرد همزمان نیتروژن و سوپرجاذب، کاربرد همزمان سوپرجاذب، اسید هیومیک و اسید سالیسیلیک، کاربرد همزمان نیتروژن، اسید هیومیک و اسید سالیسیلیک و کاربرد همزمان نیتروژن، سوپرجاذب، اسید هیومیک و اسید سالیسیلیک کارآیی مصرف آب را بهترتیب 15، 27، 32 و 40 درصد نسبت به شاهد بهبود بخشید. با توجه نتایج تجزیهی عاملی و متغیرهای متعلق به هر عامل، بهنظر میرسد عامل اول شاخصی از عملکرد محصول و عامل دوم منعکسکنندهی ویژگیهای ذاتی آرایش فضایی بوته (بار زیاد متغیرهای ارتفاع بوته و شاخص سطح برگ روی این عامل) بود. بهطور کلی، بهنظر میرسد که کاربرد همزمان نیتروژن و نهادههای بومسازگار ضمن بهبود عملکرد و خصوصیات رشدی گیاه، میتواند کارآیی مصرف آب را بهبود بخشد. همچنین انجام تجزیهی عاملی و همبستگی بالای متغیرهای درون هر عامل با یکدیگر، حاکی از آن بود که با ایجاد هر گونه تغییر در متغیرهایی که با کارآیی مصرف آب روی یک عامل قرار گرفتهاند، میتوان کارآیی مصرف آب را افزایش داده و خسارات ناشی از تنش خشکی و کمآبی را به حداقل رساند.

تأثیر سطوح مختلف خشکی در برخی صفات مورفولوژیک و فیزیولوژیک بهمنظور انتخاب متحملترین پایه انگور
چکیده:سابقه و هدف: خشکی یکی از مهمترین عوامل محدودکننده تولید محصولات کشاورزی در دنیا به شمار میرود و تابعی از ژنوتیپ، دما، بارش و ظرفیت نگهداری آب خاک است. انگور بهعنوان یکی از مهمترین محصولات باغبانی در جهان و ایران است و تنش خشکی یکی از مهمترین فاکتورهای محدودکننده رشد آن در مناطق مدیترانهای میباشد. اعمال تیمار تنش خشکی در شرایط آزمایشگاهی و انتخاب ارقام بر اساس شرایط تنش، ازجمله روشهای انتخاب ارقام متحمل به تنش خشکی میباشد که توسط سایر محققین نیز پیشنهادشده است. این پژوهش باهدف مقایسه پایه و ارقام متحمل معرفیشده توسط محققین، در شرایط تنش خشکی شدید و معرفی متحملترین انجام شد.مواد و روشها: این آزمایش با 15 تیمار شامل چهار رقم انگور غیر پیوندی (سمرقندی، یاقوتی، رطبی و چفته) و پایه پیوندی 110R که حاصل تلاقی (V. berlandieri * V. rupestris) و سه تیمار کمآبی (صفر) شاهد، 2- مگا پاسکال (تنش شدید) و 5/2- مگا پاسکال (تنش خیلی شدید) بهصورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی اجرا شد. هر واحد آزمایشی شامل یک بوته دوساله انگور گلدانی بود. صفات اندازهگیری شده در این آزمایش شامل صفات مورفولوژی (تعداد برگ سالم، طول شاخه اصلی و فرعی، سطح برگ، وزنتر و خشک ساقه و برگ) و صفات فیزیولوژی (کلروفیل کل، نشت یونی، محتوای نسبی آب برگ و میزان مالون دی آلدهید، قندهای محلول، فنل، گلایسین بتائین و اسیدآمینه پرولین) بودند.یافتهها: با افزایش شدت تنش خشکی اغلب صفات مورفولوژی مرتبط با رشد ظاهری انگور (سطح برگ، تعداد کل برگ، طول ریشه و وزنتر و خشک ساقه و ریشه) کاهش یافتند. برخی صفات فیزیولوژی (فنل، میزان کلروفیل کل و محتوای نسبی آب برگ) در مقایسه با شاهد (بدون تنش) کاهش یافتند در مقابل میزان نشت یونی، قند محلول، اسیدآمینه پرولین و گلایسین بتائین در شرایط تنش شدید و خیلی شدید افزایش یافتند. نتایج تجزیه واریانس نشان داد که اثر تنش خشکی بر تمامی صفات اندازهگیری شده بهجز وزن خشکریشه و تاج و میزان مالون دی آلدهید در پایه و ارقام متحمل این آزمایش معنیدار بود.نتیجهگیری: براساس مقایسه میانگین صفات مورفولوژی و فیزیولوژی اندازهگیری شده پس از استانداردسازی، رقم چفته بهعنوان متحملترین رقم در مقایسه با پایه و ارقام دیگر در شرایط تنش خشکی خیلی شدید معرفی میگردد و پسازآن به ترتیب ارقام رطبی، سمرقندی و یاقوتی قرار گرفتند پایه 110R به دلیل عدم همسوئی بین شاخصهای تحمل به تنش خشکی (مورفولوژی و فیزیولوژی) بهعنوان ضعیفترین پایه معرفی شد.واژههای کلیدی: پرولین، نشت یونی، سطح برگ، مالون دی آلدهید، گلایسین بتائین

ارزیابی ناسازگاری و برخی از تنظیم کنندههای رشد گیاهی بر کمیت و کیفیت میوه زالزالک قرمز (Crataegus monogyna Jacq.)
سابقه و هدف: یکی از درختان میوه معتدله که امکان پرورش آن در ایران وجود دارد، زالزالک معمولی با نام علمی Crataegus monogyna Jacq. میباشد. بهطور کلی گونههای زالزالک در مناطق معتدله نیمکره شمالی پراکنده هستند. جنس زالزالک در اکثر نقاط ایران نیز پراکندگی دارد. بیشترین پراکنش آن در شمال، شمال شرق، شمال غرب و غرب است. این جنس در ایران دارای 27 گونه و هیبرید است. زالزالک به عنوان گیاهی مقاوم به آلودگی هوا، سازگار با انواع خاکها، گیاه زینتی با ارزشی بهشمار میرود. در کشورهای اروپایی این گیاه به میزان زیادی در پرچین سازی مورد استفاده قرار میگیرد. میوه این گیاه دارای ارزش دارویی و خوراکی بوده و به اشکال مختلف مانند تازهخوری، آب میوه، مربا، ژله و به عنوان طعم دهنده غذاها مورد استفاده قرار میگیرد. اندازه کوچک و عدم یکنواختی میوههای زالزالک(Crataequs monogyna) ، محدودیتهایی برای تولید و فرآوری محصول ایجاد میکند. یکی از دلایل این محدودیت تولید گلهای فراوان و تبدیل درصد بالایی از آنها به میوه است. این آزمایش برای آگاهی از میزان سازگاری و یا ناسازگاری درخت زالزالک و همچنین تاثیر تنظیم کنندههای رشد مختلف روی میزان میوه دهی و کیفیت میوه آن انجام شد.مواد و روشها: در این پژوهش ناسازگاری احتمالی و اثر برخی تنظیمکنندههای رشد گیاهی روی خصوصیات فیزیکی و شیمیایی میوه زالزالک مورد برسی قرار گرفت. این پژوهش بصورت فاکتوریل در قالب طرح کاملاً تصادفی با عوامل گرده افشانی در دو سطح (خود گردهافشان و آزاد گردهافشان) و تنظیم کنندههای رشد جیبرلیک اسید (75 و 150 ppm)، نفتالین استیک اسید (10 و 20 ppm) و اتفون (75 و 150 ppm) در سه تکرار به اجرا در آمد. شاخصهای درصد تشکیل میوه (3، 7، 10 و 17 هفته پس از تمام گل) وزن دانه، گوشت و میوه، نسبت وزن گوشت به بذر، وزن خشک و درصد آب گوشت، TSS، TA، TSS/TA و تعداد سلول در هر میدان دید میکروسکوپ براورد گردید.یافته ها: نوع گرده افشانی (خود و آزاد گرده افشان) و همچنین محلول پاشی تنظیم کنندههای رشد گیاهی بر درصد تشکیل میوه زالزالک، وزن میوه، وزن دانه و نسبت گوشت به دانه موثر بود در حالی که تیمارهای انجام شده در وزن گوشت و درصد آب میوه تاثیر معنیداری نداشت. براساس نتایج بهدست آمده خصوصیات کیفی میوه زالزالک (مواد جامد محلول، اسید کل و نسبت مواد جامد محلول به اسید کل) تحت تاثیر تنظیم کنندههای رشد گیاهی مورد استفاده در این آزمایش (GA، NAAو ETHE) قرار گرفتند همچنین تعداد سلولها تحت تاثیر تیمارها قرار گرفت بهطوری که اتفون در غلظتهای 75 و 150 ppm بیشترین تاثیر را بر تعداد سلول در درختان آزاد گرده افشان و خود گردهافشان داشت.نتیجه گیری: نتایج نشان داد که زالزالک استفاده شده در این آزمایش دارای ناسازگاری است و برای تشکیل میوه بهتر نیاز به گرده افشانی آزاد (استفاده از گرده افشان) دارد. همچنین استفاده از تنظیم کننده های رشد گیاهی بر درصد تشکیل میوه و اندازه میوه موثر بودند. بیشترین درصد تشکیل میوه با محلول پاشی جیبرلین حاصل شد و بزرگترین میوه ها حاصل از محلول پاشی با اتفن بود.واژههای کلیدی: اتفون، جیبرلین، زالزالک، گرده افشانی، نفتالین استیک اسید

مطالعه خصوصیات مورفولوژیکی ژنوتیپهای مختلف زرشک (Berberis. sp) در منطقه شمال شرق ایران
سابقه و هدف: ایران با قرار داشتن در زمرة مناطق اصلی تنوع گیاهی ذخایر ژنتیکی مطلوبی در زمینهی گیاهان به ویژه محصولات باغبانی دارد. زرشک نیز از جمله ریزمیوههایی است که به دلیل وجود جمعیتهای وحشی در سراسر کشور مستعد انجام پژوهشهای اولیه شامل گردآوری ژنوتیپهای مختلف و بررسی آنها در جهت شناسایی مزایا و معایب این گیاه و در نهایت گروهبندی آنها میباشد. هدف از اجرای این تحقیق بررسی خصوصیات مورفولوژیکی ژنوتیپهای مختلف زرشک شمال غرب ایران و بررسی قرابتهای آنها با هم می باشد.مواد و روشها: این مطالعه به منظور بررسی خصوصیات مورفولوژیکی ژنوتیپهای مختلف زرشک در منطقه شمال غربی ایران در سال 1396 اجرا شد. در این مطالعه از استانهای خراسان شمالی، خراسان جنوبی، خراسان رضوی و گلستان 15 ژنوتیپ مختلف زرشک جمعآوری شد و خصوصیات مورفولوژیکی آنها از قبیل خصوصیات مربوط به برگ و میوه ژنوتیپهای زرشک مورد بررسی قرار گرفت و در پایان همبستگی آنها انجام و تجزیه کلاستر نیز صورت گرفت. یافتهها: نتایج نشان داد که اثر تیمار ژنوتیپ بر تمامی صفات بررسی شده در این مطالعه معنی دار بود. با توجه به این نتایج مشخص شد که تعداد برگ در هر گره (6/8 برگ)، طول خار (96/3 سانتی متر) و زاویه خار (106 درجه) در ژنوتیپ درگز2 بیشتر از سایر ژنوتیپهای مورد مطالعه بود در حالی که طول دمبرگ (42/1 سانتی متر) در ژنوتیپ شیروان2 بالاتر از سایر ژنوتیپها بود. با توجه به اینکه طول خوشه (3/7 سانتی متر) و وزن خشک صد حبه (55/8 گرم) ژنوتیپ شیروان3 بیشتر از سایر ژنوتیپها بود ولی نتایج نشان داد که ژنوتیپ بی دانه بیرجند از نظر صفت تعداد برگ در هر خوشه (18 برگ در هر خوشه) و طول دم میوه (6/9 میلی متر) برتر از سایر ژنوتیپهای دانه دار بود. همچنین نتایج نشان داد ژنوتیپ گلستان5 از نظر صفات طول حبه (97/11 میلی متر) و نسبت طول به قطر حبه (98/1) نسبت به سایر ژنوتیپ ها برتر بود. صفات وزن تر و خشک و حجم صد حبه با صفات طول خوشه، طول حبه، قطر حبه مثبت بود وبا افزایش این صفات وزن تر و خشک حبه در ژنوتیپهای زرشک افزایش یافت. نتایج حاصل از تجزیه کلاستر در فاصله 5/0 ژنوتیپهای زرشک را در 5 گروه اصلی گروه بندی نمود که در گروه اول ژنوتیپ شیروان3 و در گروه پنجم ژنوتیپ درگز1 قرار داشت که نشان دهنده اختلاف این ژنوتیپ ها با سایر ژنوتیپ های مورد مطالعه می باشد. همچنین ژنوتیپ بیرجند بی دانه به همراه ژنوتیپ های شیروان1، شیروان4، شیروان2، درگز3 و درگز2 در یک گروه قرار داشتند که بیانگر نزدیک بودن ویژگی های مورفولوژیکی این ژنوتیپ ها با هم می باشد. نتیجهگیری: نتایج حاصل از این تحقیق نشان داد که ژنوتیپهای مختلف زرشک دارای طیف وسیعی از خصوصیات مورفولوژیکی بودند که با استفاده از اشتراکاتی که دارند آنها را در گروههای مشابه تقسیم بندی نموده و میتوان از این اطلاعات برای اهداف اصلاحی ژنوتیپهای زرشک استفاده نمود.

اثر تاریخ کاشت و تراکم بر سرعت تولید و زوال برگ گلرنگ
مقدمهپیشبینی تغییرات سطح برگ بخش مهمی از مدلهای شبیهسازی گیاهان زراعی است. قابلیت پیش بینی تغییرات شاخص سطح برگ گیاه در طی فصل رشد برای تخمین میزان تشعشع خورشیدی دریافتی و ماده خشک تولیدی گیاه مهم است. بنابراین هدف از این مطالعه بررسی روند تغییرات تولید و زوال سطح برگ گلرنگ "توده محلی اصفهان" میباشد.مواد و روشیک آزمایش فاکتوریل در قالب طرح بلوکهای کامل تصادفی در 4 تکرار در مزرعه تحقیقاتی دانشگاه ولیعصر در سال 1391 انجام شد. عوامل آزمایش شامل تاریخ کاشت (17 فروردین و 6 اردیبهشت) و تراکم بوته (15، 40، 65 و 90 بوته در مترمربع) بودند. پس از سبز شدن بوتهها تا پایان فصل رشد به فاصله زمانی هر 10 روز یکبار سطح برگ بوتهها مورد اندازهگیری قرار گرفت. همچنین از ابتدای فصل رشد تا پایان مرحله گلدهی، هر 7-3 روز یکبار تعداد برگ سبز و گره تولید شده روی ساقه اصلی، تعداد برگ زرد روی ساقه اصلی و همچنین تعداد کل برگ زرد روی بوته شمارش و ثبت گردید.یافتههانتایج نشان داد با کاهش تراکم شاخص سطح برگ حداکثر (LAImax) روند افزایشی داشت بهطوری که مقدار آن در تراکم 15 بوته در مترمربع به 91/3 و 37/3 در تاریخ کاشتهای اول و دوم رسید و در تراکم های بالاتر مقدار آن کاهش یافت. سرعت تولید برگ (leaf/°Cd-1) میان تراکمهای مختلف در تاریخ کاشت های مورد مطالعه اختلاف معنیداری نشان نداد و بین 73/1 تا 93/1 leaf/°Cd-1 متغیر بود. تولید برگ در گیاه پس از گذشت 1349 تا 1395 °Cd-1 و 1300 تا 1384 °Cd-1 بهترتیب برای تاریخ کاشتهای اول و دوم پایان یافت. با افزایش تعداد برگ در طول فصل رشد، سطح برگ بهصورت توانی افزایش یافت بهطوری که شیب افزایش سطح برگ با افزایش تراکم و تاخیر در کاشت روند کاهشی داشت. سرعت زوال برگ در تراکمهای پایینتر هر تاریخ کاشت و نیز با تأخیر در کاشت با شیب زیادی صورت گرفت. سرعت پیری برگ براساس تعداد برگ زرد در بوته بین 0025/0 تا 0045/0 leaf/°Cd-1 برای تاریخ کاشت اول و 0018/0 تا 0037/0 leaf/°Cd-1 برای تاریخ کاشت دوم متغیر بود.بحثنتایج نشان داد با افزایش تراکم از شاخص سطح برگ حداکثر کاسته شد. با افزایش تراکم بوته به دلیل افزایش رقابت درون و برون بوتهای و درنتیجه کاهش تعداد شاخه فرعی در هر بوته، سطح برگ تک بوته کاهش مییابد اما بر خلاف این انتظار افزایش تعداد بوته نتوانست کاهش شاخص سطح برگ را جبران کند. این امر احتمالا به دلیل کاهش تعداد برگ در بوته در تراکم بالاتر بود. تأخیر در کاشت موجب افزایش سرعت ظهور برگ و کاهش دوره موثر تولید برگ گردیده و از تعداد نهایی برگ در ساقه اصلی کاسته شد. سرعت تولید گره در طی این دوره در درجه نخست به وسیله دما و پس از آن توسط دسترسی به اسمیلات برای رشد برگ تعیین میشود. با افزایش تراکم، سرعت زوال برگ در ساقه بیشتر شد. افزایش شدت پیری در تراکمهای کاشت پایین را میتوان به رقابت درون گیاهی و سایهاندازی بیشتر نسبت داد.

ارزیابی پاسخ به تنش خشکی در برخی از ژنوتیپهای امید بخش و ارقام ایرانی و خارجی زیتون با استفاده از شاخص-های مبتنی بر عملکردهای میوه و روغن
سابقه و هدف: زیتون درختی همیشه سبز و یکی از قدیمیترین گیاهان کشت شده در حوزه مدیترانه شناخته شده است. تنش خشکی به عنوان یکی از مهمترین تنش غیرزیستی سبب کاهش عملکرد زیتون در شرایط گرمسیری و نیمهگرمسیری میباشد. بنابراین شناسایی ارقام مقاوم به تنش جهت کشت در مناطق مستعد خشکی یکی از کارهای مهم در توسعه سطح زیر کشت این گیاه باغی محسوب میشود. این آزمایش با هدف ارزیابی برخی از ارقام تجاری و ژنوتیپهای امیدبخش زیتون با استفاده از شاخصهای تحمل و حساسیت به تنش خشکی صورت گرفت.مواد و روشها: بهمنظور بررسی اثر تنش خشکی بر عملکرد میوه و درصد روغن 20 رقم تجاری و ژنوتیپ امیدبخش زیتون یک آزمایش فاکتوریل در قالب طرح بلوکهای کامل تصادفی با سه تکرار در ایستگاه تحقیقات زیتون طارم طی دو سال باغی 1396-1394 اجرا شد. اعمال تنش خشکی به صورت قطع آبیاری بهمدت یک ماه قبل از سخت شدن تا زمان سخت شدن هسته صورت گرفت. محاسبه 9 شاخص تحمل و حساسیت به تنش خشکی بر اساس عملکرد میوه و درصد روغن در شرایط تنش و بدون تنش خشکی، محاسبه همبستگی بین عملکرد و شاخصها و نیز تجزیه به مؤلفههای اصلی با استفاده از ماتریس ضرایب همبستگی انجام شد. یافتهها: بر اساس نتایج به دست آمده تنش خشکی موجب کاهش عملکرد میوه در کلیه ژنوتیپها و ارقام و افزایش درصد روغن در برخی از آنها شد. نتایج تجزیه همبستگی نشان داد کلیه شاخصها به جز TOL و SSI دارای همبستگی مثبت و معنیداری با عملکرد میوه و درصد روغن در شرایط تنش و بدون تنش خشکی هستند. نتایج تجزیه به مؤلفههای اصلی (PCA) نشان داد دو مؤلفه نخست در مجموع 78/98 و 59/99 درصد کل تغییرات مربوط به عملکرد میوه و درصد روغن را توجیه نمودند. زاویه بین بردارهای هر یک از شاخصها در بایپلات ترسیم شده توسط دو مؤلفه نخست نتایج تجزیه همبستگی را تأیید نمود. با توجه به نمودار سهبعدی ترسیم شده بر اساس مقادیر عملکرد و شاخص STI مشخص شد که ژنوتیپ امیدبخش T7 در هر سال باغی و متوسط دو سال از نظر عملکرد میوه و درصد روغن در گروه A قرار داشت. با در نظر گرفتن نتایج تجزیه به مؤلفههای اصلی نیز این ژنوتیپ در زمره ارقام برتر قرار گرفت.نتیجهگیری: نتایج به دست آمده از این آزمایش نشان داد ژنوتیپ امیدبخش T7 نسبت به سایر ارقام و ژنوتیپها به عنوان متحملترین ژنوتیپ به تنش خشکی شناسایی شد. لذا استفاده از آن در برنامههای اصلاحی ارقام متحمل و همچنین کشت در مناطق مستعد خشکی توصیه میشود.

اثر محلولپاشی تغذیه نیتروژن و اتفون بر گلانگیزی و برخی از خصوصیات فتوسنتزی خیار ویکتور
سابقه و هدف: میوهها و سبزیها بخش مهمی از رژیم غذایی انسان را تشکیل میدهند که در این میان خیار دارای اهمیت ویژهای است. خیارهای مزرعهای معمولا گل نر بیشتری نسبت گل ماده تولید میکنند. اهمیت تولید گل ماده در تشکیل میوه و رابطه مستقیم آن با عملکرد محصول دلیل مطالعه عوامل تاثیرگذار بر تحریک و تولید گل ماده می باشد لذا هدف از این پژوهش بررسی نقش هورمون اتیلن و تغذیه نیتروژن و اثر متقابل آنها بر آرایش گلدهی و جنسیت گلهای تولیدی خیار بود. همچنین تاثیرگذاری تیمار اتفون و تغذیه نیتروژن بر شاخصهای فتوسنتزی مورد بررسی قرار گرفت تا سازوکارهای فیزیولوژیکی اثر این دو تیمار بر گلدهی روشن شود.مواد و روشها: آزمایش بهصورت کرتهای خرد شده در قالب طرح کاملا تصادفی و کشت گلدانی در گلخانه با سه تکرار و 6 تیمار در دانشگاه علوم کشاورزی و منابع طبیعی ساری در سال 1396-1397 انجام پذیرفت. کرت اصلی شامل اتفون در دو سطح صفر و 5/0 میلیمولار و کرت فرعی شامل اوره در سه سطح صفر، 2 و 4 گرم در لیتر هر کدام با سه مرحله محلولپاشی بود. اولین محلولپاشی با اوره در مرحله دو برگی شروع گردیده و سه روز بعد از آن محلولپاشی با اتفون انجام شد. مرحله دوم پاشش اوره پنج روز بعد از پاشش اول آن انجام گردید و مجددا سه روز بعد از مرحله دوم اوره تیماره اتفون تکرار شد و این روند برای مرحله سوم هم اجرا شد. در این تحقیق اثر تیمارهای اتفون و کود نیتروژن بر خصوصیات فیزیولوژیک و گل انگیزی مطالعه و خصوصیاتی شامل تعداد گلهای نر و ماده همراه با مورفولوژی گل مانند قطر تخمدان، طول جام گل و طول مادگی و پارامترهای تبادل گازی مانند سرعت تعرق، نرخ جذب دیاکسید کربن، هدایت روزنهای ارزیابی شدند.یافتهها: بیشترین تعداد گل نر در تیمارهای شاهد (3/44) و اوره دو در هزار (44) بدون کاربرد اتفون دیده شد. همچنین بیشترین تعداد گل ماده متعلق به تیمار حاوی اوره چهار در هزار بدون کاربرد اتفون (24) و اتفون نیم میلیمولار بدون اوره (6/17) بود. علاوه بر این بیشترین طول مادگی متعلق به محلولپاشی اوره چهار در هزار بدون کاربرد اتفون با میانگین 6/4 میلیمتر بود که اختلاف معنیداری با اوره دو در هزار با کاربرد اتفون نیم میلیمولار (2/4 میلیمتر) نداشت. بیشترین سرعت تعرق در تیمارهای اوره دو در هزار بدون اتفون (54/3 میلیمول بر مترمربع در ثانیه) و اتفون نیم میلیمولار بدون کاربرد اوره (66/3 میلیمول بر مترمربع در ثانیه) دیده شد که نسبت به شاهد برتر بودند. تیمار حاوی اتفون نیم میلیمولار بدون کاربرد اوره (9/241 میلیمول بر مترمربع در ثانیه) همچنین بیشترین هدایت روزنهای را نشان داد که با سایر تیمارهای اتفونی و اوره دو در هزار بدون اتفون اختلاف معنیداری نداشت. بیشترین میزان جذب دیاکسید کربن متعلق به تیمار اتفون با اوره چهار در هزار (6/8 میکرومول بر مترمربع در ثانیه) بود که نسبت به شاهد برتری نشان داد. نتیجهگیری کلی: در مجموع میتوان نتیجه گرفت که اثر متقابل تیمارهای نیتروژن و اتفون در اکثر فاکتورهای اندازهگیری شده زایشی و رویشی موجب بهبود پارامترهای مورد مطالعه شد. از آنجایی که یکی از اهداف پژوهش افزایش تعداد گل ماده است بنابراین دو تیمار اوره چهار گرم در هزار بدون اتفون و اتفون نیم میلیمولار بدون کاربرد اوره میتوانند تاثیر مثبتی بر تعداد گل ماده در گیاه خیار داشته باشند.

وراثت پذیری و هتروزیس برخی صفات مرتبط با ارزش غذایی و کیفیت فیزیکی دانه برنج
سابقه و هدف: برنج غذای اصلی مردم ایران و بسیاری از کشورهای جهان میباشد. با توجه به افزایش جمعیت و وجود محدودیتهای اقلیمی برای کشت ارقام زراعی مهم از جمله برنج، لازم است که تولید برنج و در کنار آن ارزش غذایی افزایش یابد. بهرهبرداری از هتروزیس به عنوان راهکاری برای افزایش بیشتر عملکرد و کیفیت دانه برنج میباشد. بنابراین، هدف از این مطالعه، بررسی ساختار ژنتیکی و تعیین هتروزیس صفات مورد مطالعه برای برنامههای اصلاحی و انتخاب ترکیبهای مناسب برای تولید برنج هیبرید میباشد.مواد و روشها: در این آزمایش از 24 هیبرید حاصل از تلاقی شش لاین در چهار تستر (که براساس برخی صفات مهم غذایی مورد دورگگیری قرار گرفته بودند) استفاده گردید. هیبریدها و والدین در قالب طرح بلوکهای کامل تصادفی در سه تکرار براساس پنج صفت ارزش غذایی (فعالیت آنتیاکسیدانی، محتوای فنل، عناصر آهن و روی و پروتئین) و هفت صفت کیفیت فیزیکی (درصد برنج سالم و خرد، راندمان تبدیل، درصد گچیبودن دانه، طول، عرض و شکل دانه) به همراه عملکرد براساس طرح تلاقی لاین × تستر مورد ارزیابی قرار گرفتند و اثرات ترکیبپذیری والدین و هیبریدها، وراثتپذیری عمومی و خصوصی و هتروزیس صفات برآورد گردید.یافتهها: نتایج تجزیه واریانس نشان داد که اثر متقابل لاین × تستر برای صفات معنیدار بود که این حاکی از وجود اثر غیرافزایشی ژن در کنترل صفات میباشد. با توجه به نتایج ترکیبپذیری عمومی، ارقام فجر و غریب به عنوان بهترین ترکیبشونده برای عملکرد و صفات ارزش غذایی شناخته شدند. اجزای واریانس ژنتیکی برای صفات مورد بررسی نشان از سهم بیشتر واریانس ترکیبپذیری خصوصی نسبت به واریانس ترکیبپذیری عمومی بود و این نشاندهنده اثر غیرافزایشی ژن در وراثتپذیری صفات است. در تحقیق حاضر، تمامی صفات وراثتپذیری عمومی بالا (بالای 95 درصد) و خصوصی پایین داشتند. بررسی پدیده هتروزیس در تلاقی مورد بررسی سهم واریانس غالبیت را در کنترل این صفات نشان داد. نتیجهگیری: نتایج حاصل از بررسی اجزای واریانس ژنتیکی و بررسی هتروزیس نشان از نقش غیر افزایشی ژن در کنترل صفات ارزش غذایی، فیزیکی و عملکرد میباشد. بنابراین، بهترین روش اصلاحی استفاده از هتروزیس و روش هیبریداسیون در این تحقیق میباشد. نتایج هتروزیس نشان داد که برای اصلاح همزمان صفاتی مثل آهن، روی و پروتئین به همراه افزایش درصد برنج سالم، راندمان تبدیل، طول دانه و عملکرد و کاهش درصد گچیبودن و درصد برنج خرد میتوان از تلاقیهای اوندا × سپیدرود، غریب × آمل2، فجر × آمل2، فجر × آبجیبوجی، اوندا × موسیطارم و اوندا × صدری استفاده نمود.

اثر محلولپاشی برگی سالیسیلیکاسید بر ویژگیهای مورفوفیزیولوژیک و بیوشیمیایی گیاه فیسالیس (Physalis peruviana L.) در شرایط تنش شوری
سابقه و هدف: ﺷﻮری ﺧﺎک از مهمترین ﻣﺸﻜﻼت ﻛﺸﺎورزی در ﻣﻨﺎﻃﻖ ﺧﺸﻚ و نیمه ﺧﺸﻚ دﻧﻴﺎ میباشد. تنش شوری از طریق برهم زدن توازن عناصر موجود در خاک، موجب استقرار ضعیف گیاه میشود، که میزان این تأثیر بستگی به میزان حساسیت گیاه دارد. از طرف دیگر سالیسیلیکاسید، بهعنوان یک تنظیمکننده رشد درونی، یک ترکیب موثر در سیستم دفاعی گیاهان محسوب میشود. فیسالیس (Physalis peruviana L.) متعلق به خانواده سولاناسه میباشد. پراکندگی جغرافیایی این گیاه بیشتر در مناطق گرمسیر آمریکای جنوبی و شرق آسیا بوده و در دامنه وسیعی از شرایط آب و هوایی رشد میکند. این تحقیق با هدف بررسی اثر اﺳﻴﺪ ﺳﺎﻟﻴﺴﻴﻠﻴﻚ بر برخی صفات مورفولوژیکی و بیوشیمیایی ﮔﻴﺎه فیسالیس در شرایط ﺗﻨﺶ ﺷﻮری انجام شد. مواد و روشها: این آزمایش در سال 1397 در گلخانه تحقیقاتی دانشکده کشاورزی دانشگاه لرستان بهصورت فاکتوریل، در قالب طرح کاملا تصادفی، با سه تکرار انجام شد. فاکتورهای آزمایش، شامل چهارسطح شوری (0 ، 35، 70 و 105میلیمولار کلرید سدیم) و چهار سطح سالیسیلیکاسید (0، 5/0، 1 و 2 میلیمولار) بودند. نشاهای فیسالیس در گلدان حاوی خاک، کود دامی و ماسه به نسبت 2:1:1 کشت شدند. صفات اندازهگیری شده در این آزمایش شامل ارتفاع بوته، قطر ساقه، تعداد و سطح برگ، وزن تر و خشک ریشه، برگ، ساقه و کل شاخساره، کلروفیل، آنتوسیانین، کارتنویید، پرولین، نشت یونی، مالون دی آلدهید و محتوای نسبی آب برگ بودند.یافتهها: نتایج تجزیه واریانس دادهها نشان داد، اثرات اصلی و اثرات متقابل تنش شوری و سالسیلیکاسید برای اکثر صفات اندازهگیری شده معنیدار شد. با افزایش غلظت کلریدسدیم، ارتفاع گیاه، ﻗﻄﺮ ﺳﺎﻗﻪ، ﺗﻌﺪاد ﺑﺮگ، وزن تر و ﺧﺸﻚ ﺷﺎﺧﺴﺎره و رﻳﺸﻪ، میزان کلروفیل و کارتنویید، آنتوسیانین برگ، محتوای نسبی و سطح برگ، کاهش، و میزان مالون دی آلدهید، نشت الکترولیت و پرولین افزایش یافت و کاربرد سالیسیلیکاسید موجب کاهش اثرات منفی ناشی از تنش شوری شد، بهطوریکه غلظت 2 میلیمولار سالیسیلیکاسید بیشترین اثر را بر صفات مورد بررسی داشت، در بین تیمارها، بیشترین میانگین برای اکثر صفات در تیمار شاهد (بدون تنش شوری) و غلظت 2 میلیمولار سالیسیلیکاسید مشاهد شد. بیشترین (09/79 درصد) و کمترین (59/59 درصد) میزان محتوای نسبی به ترتیب در تیمار بدون تنش شوری (با کاربرد غلظت 2 میلیمولار سالیسیلیکاسید) و تیمار شوری 105 میلیمولار کلریدسدیم (با غلظت صفر میلیمولار سالیسیلیکاسید) مشاهده شد. بیشترین میزان مالوندیآلدهید در تیمار تنش شوری 105 میلیمولار، غلظت صفر میلیمولار سالیسیلیکاسید مشاهده شد.نتیجهگیری: ﻧﺘﺎﯾﺞ به دست آمده از پژوهش، نشان داد که اﻓﺰاﯾﺶ غلظت کلرید سدیم، ﺳـﺒﺐ ﮐـﺎﻫﺶ ﻣﺤﺘـﻮای ﻧﺴـﺒﯽ آب ﺑـﺮگ، رنگدانههای فتوسنتزی، وزن تر و خشک گیاه و سایر صفات مورفوفیزیولوژیک و بیوشیمیایی فیسالیس شد و کاربرد برگی سالیسیلیکاسید، موجب بهبود این صفات شد. به نظر میرسد گیاه فیسالیس، گیاهی حساس به شوری است و شوری بیش از 35 میلیمولار را تحمل نمیکند، اما کاربرد سالیسیلیکاسید در غلظت 2 میلیمولار میتواند تا حدودی اثرات منفی ناشی از تنش شوری را کاهش دهد. با توجه به نتایج حاصل از پژوهش، گیاه فیسالیس برای کشت در مناطق دارای آب شور توصیه نمیشود.

مطالعه کشت مخلوط ردیفی ژنوتیپهادر گندم دوروم(Triticum durum) در شرایط تنش کم آبی
سابقه و هدف: گندم یکی از مهمترین محصولات غذایی در جهان است که رشد و تولید آن اغلب با تنش خشکی کاهش مییابد. یکی از روشهای ممکن افزایش غذا به روش پایدار با حفظ سطح زیر کشت استفاده از سیستم کشت مخلوط است. کشت مخلوط به عنوان یکی از روشها و نمونهای از نظامهای پایدار در کشاورزی اهدافی نظیر ایجاد تعادل اکولوژیک، بهرهبرداری بیشتر از منابع، افزایش کمی وکیفی عملکرد، کاهش خسارت آفات بیماریها و علفهای هرز را دنبال میکند. کاهش وابستگی کشاورزان به آفتکشها به شرط حفظ کیفیت محصول و بازار پسندی آن یکی از اهداف کشت مخلوط در کشاورزی پایدار است .بطورکلی هدف از اجرای این آزمایش ارزیابی اثر تنوع ژنتیکی ژنوتیپها بر عملکرد و اجزاء عملکرد دانه تحت شرایط تنش کم آبی بعد از گلدهی در سیستمهای کشت مخلوط گندم دوروم می باشد. مواد و روشها: این آزمایش بصورت اسپلیت پلات در قالب طرح بلوکهای کامل تصادفی با سه تکرار در سال زراعی 97-96 در دانشکده کشاورزی و منابع طبیعی داراب، دانشگاه شیراز انجام شد. عاملها شامل: رژیم رطوبتی بعد از گلدهی در دو سطح (آبیاری مطلوب و تنش کم آبی) و سیستمهای کاشت شامل کشت خالص ژنوتیپهای گندم دوروم (کشت خالص لاین 4-92DW-، کشت خالص شبرنگ، کشت خالص لاین 14-94DW-، کشت خالص بهرنگ) و کشت مخلوط آنها (کشت مخلوط لاین 4-92 DW-+ شبرنگ، کشت مخلوط لاین 4-92 DW-+ 14-94 DW-، کشت مخلوط 4-92 DW-+ بهرنگ، کشت مخلوط شبرنگ + 14-94 DW-، کشت مخلوط شبرنگ + بهرنگ، کشت مخلوط 14-94 DW-+ بهرنگ، کشت مخلوط 4-92 DW-+ شبرنگ + 14-94 DW-+ بهرنگ) بود. یافتهها: بر طبق نتایج این آزمایش بیشترین عملکرد دانه در تیمار کشت مخلوط دو ژنوتیپ بهرنگ+ 14-94DW- با میانگین 8815 کیلوگرم در هکتار در شرایط آبیاری مطلوب به دست آمد که تفاوت معنیداری با مخلوط چهار ژنوتیپ در شرایط آبیاری مطلوب نداشت. کمترین عملکرد دانه در تیمار کشتهای مخلوط بهرنگ + 4-92 DW-با مقدار 2233 کیلوگرم در هکتار در شرایط تنش کم آبی مشاهده شد. همچنین مقایسه گروهی کشتهای خالص و کشت مخلوط دوتایی و چهارتایی این ژنوتیپ ها نشان داد که بیشترین عملکرد دانه در تیمار کشت مخلوط چهارتایی تحت شرایط آبیاری مطلوب با میانگین 8799 کیلوگرم در هکتار بدست آمد. تجزیه ضرایب همبستگی در این آزمایش نشان داد که در شرایط تنش کم آبی حداکثر همبستگی مثبت و معنیدار بین عملکرد دانه و عملکرد بیولوژیک (**76/0=r) مشاهده گردید. همچنین در شرایط آبیاری مطلوب بیشترین همبستگی مثبت و معنیدار بین عملکرد دانه با عملکرد بیولوژیک (**90/0=r) وجود داشت.نتیجهگیری: استفاده از کشت مخلوط ژنوتیپهای مختلف گندم دوروم، عملکرد و اجزای عملکرد را نیز تحت تاثیر قرار داد. کشت مخلوط ژنوتیپها در شرایط آبیاری مطلوب بیشترین عملکرد را نسبت به شرایط تنش کم آبی به خود اختصاص داد. در مجموع به نظر میرسد کشتهای مخلوط به دلیل داشتن پتانسیل عملکرد دانه بالاتر چه در شرایط آبیاری مطلوب و چه در شرایط محدودیتهای رطوبتی قابل توصیه میباشند.

تأثیر طیفهای نور و تنظیم کننده رشد تیدیازورون بر رویانزایی و سیستم فتوسنتزی ارکیده فالانوپسیس
سابقه و هدف: فالانوپسیس (Phalaenopsis amabilis) از انواع ارکیده های مناطق گرمسیری است که در سالیان اخیر به عنوان گیاه گلدانی و گل شاخه بریده در بازار ایران بسیار مورد توجه قرار گرفته است. به دلیل تخصصی بودن روش کشت و عدم تولید نشاء این گیاه در داخل کشور، پرورش دهندگان داخلی مجبور به واردات گیاهچه های فالانوپسیس از کشورهای دیگر هستند. با وجود مشکلات فراوان از قبیل تلفات حمل و نقل، قرنطینه سخت گیرانه و زمان بر، قیمت و هزینه های تمام شده بالا در تولید، واردات این نشاها همواره روند افزایشی داشته است. معرفی ارقام جدید با کیفیت بالا، زیبایی و جذابیت برای تامین نیاز بازار جهانی، منجر به تقویت و تشدید این وابستگی دائمی شده است. پیدا کردن راهی برای تکثیر ارقام این گیاه اولین گام برای از بین بردن این وابستگی است. بنابراین هدف از این تحقیق ارزیابی اثرات طیف نور، تنظیم کننده های رشد گیاهی و اثر زخم زنی برای یافتن شرایط مناسب به منظور انگیزش جنین زایی بدنی و تولید انبوه کلون این گیاه بود. مواد و روش ها: برای انجام پژوهش پس از کشت بذر از پروتوکورم های 3 ماهه درون شیشه ای به عنوان ریز نمونه استفاده شد که به دو شکل برش زده و بدون برش روی محیط کشت 1/2 MS با غلظت های صفر و 3 میلی گرم در لیتر تیدیازورن کشت شدند و طی چهار ماه تحت تأثیر 6 نوع طیف نوری با استفاده از لامپ های ال ای دی شامل طیف های نوری آبی، قرمز، سفید، سبز، آبی- قرمز، قرمز- قرمز دور و همچنین تاریکی قرار گرفتند. واکشت ریز نمونه ها هر بیست روز یک بار تکرار شد. دوبار تصویر برداری فلورسنس (ابتدا دو ماه پس از القاء تیمار و سپس در پایان تیمارهای رویان زایی) برای ارزیابی حداکثر کارآیی فتوسیستم دو (QY-max) انجام شد. یافته ها: نتایج نشان داد که طیف های مختلف نوری و تنظیم کننده رشد می توانند روی القاء و تشکیل رویان های بدنی بسیار موثر باشند، اما زخم زنی بر رویان زایی اثر نداشته است. پروتوکورم های بدون برش در تیمار دارای غلظت 3 میلی گرم در لیتر تیدیازورن و طیف نوری قرمز- قرمز دور بدون اختلاف معنی داری با طیف قرمز بالاترین درصد رویان زایی بدنی مستقیم (100%) را داشتند. کمترین میزان رویان زایی به ترتیب در طیف های آبی، تاریکی و آبی قرمز مشاهده شد. بالاترین میزان QY-max در تصویربرداری اول در تیمار نور آبی و بالاترین میزان QY-max در تصویربرداری دوم در تیمار نوری سفید و سبز مشاهده گردید.نتیجه گیری: در مجموع استفاده از طیف نوری قرمز- قرمز دور در کنار استفاده از تنظیم کننده رشد گیاهی تیدیازورن برای القاء موفقیت آمیز رویان بدنی در این نوع ارکیده توصیه می شود. بر اساس اندازه گیری فلورسنس کلروفیل جنین های بدنی می توان گفت پس از القاء رویان، استفاده از نور آبی و در ادامه نور سفید یا سبز می تواند برای رسیدن به حداکثر کارایی سیستم فتوسنتزی مورد استفاده قرار گیرد.

سابقه و هدف: گل نرگس یکی از مهمترین گیاهان زینتی و دارویی است که گونههای مختلف آن در سرتاسر دنیا به جز مناطق گرمسیری رشد میکنند. نرگس شهلا گیاهی سوخدار و چندساله است که از آن به عنوان گل شاخه بریده، باغچهای و گلدانی استفاده میشود. با توجه به اینکه گل نرگس شهلا یکی از محصولات مهم اقتصادی و زیر کشت در ایران است و از طرف دیگر بحران خشکسالی و شوری آب و خاک از مشکلات جدی بخش تولید در کشاورزی است، آگاهی از میزان تحمل این گیاه به تنشهای خشکی و شوری به منظور تولید بهینه محصول، امری ضروری است. این پژوهش با هدف مطالعه تأثیر توأم تنشهای خشکی و شوری بر برخی از خصوصیات زایشی، بیوشیمیایی و غلظت یونهای سدیم و پتاسیم گل نرگس انجام شد.مواد و روشها: این پژوهش گلدانی به صورت فاکتوریل در قالب طرح کاملا تصادفی با سه تکرار انجام شد. فاکتور اول تنش خشکی در چهار سطح 90 (شاهد)، 70، 50 و 30 درصد ظرفیت زراعی و فاکتور دوم تنش شوری آب آبیاری ناشی از کلرید سدیم در چهار سطح صفر (شاهد)،20، 40 و 60 میلیمولار بودند. اعمال تیمارهای خشکی و شوری حدود 4 ماه به طول انجامید و سپس صفات مورد نظر اندازهگیری شدند. صفات مورد بررسی شامل تعداد گل، قطر گل، کلروفیل کل، کاروتنوئید و فلاونوئید برگ، آنزیمهای کاتالاز و گایاکول پراکسیداز برگ و مقادیر عناصر سدیم و پتاسیم برگ و سوخ بودند.یافتهها: نتایج نشان داد اثر تنشهای خشکی و شوری و اثر متقابل آنها بر تعداد گل معنیدار نبود؛ ولی این تنشها سبب کاهش قطر گل شدند. بیشترین و کمترین قطر گل به ترتیب از تیمارهای شاهد و تنش خشکی 30 درصد ظرفیت زراعی همراه با شوری 60 میلیمولار بدست آمد. اثر متقابل تنشهای شوری و خشکی بر محتوای کلروفیل کل معنیدار و کاهشی بود. بهطوریکه تیمار 60 میلیمولار کلرید سدیم همراه با 30 درصد ظرفیت زراعی باعث کاهش 72 درصدی کلروفیل کل نسبت به شاهد شد. تنشهای شوری و خشکی سبب کاهش محتوای کاروتنوئید برگ شدند، بهطوریکه مقدار این صفت در شدیدترین سطوح شوری و خشکی، به ترتیب 26 و 25 درصد در مقایسه با شاهد کاهش یافت. نتایج نشان داد که با افزایش سطوح تنش شوری و خشکی میزان فعالیت آنزیمهای کاتالاز و گایاکول پراکسیداز افزایش یافت، بهطوریکه بیشترین فعالیت آنزیمی از بالاترین سطوح (شوری 60 میلیمولار و 30 درصد ظرفیت مزرعه) بدست آمد. در اثر متقابل دو تنش، بیشترین میزان فعالیت آنزیم کاتالاز مربوط به تیمار 60 میلیمولار همراه با 30 درصد ظرفیت زراعی با افزایش 5/4 برابری نسبت به شاهد بدست آمد. با افزایش تنش شوری و خشکی میزان پتاسیم برگ و سوخ کاهش یافت، ولی سدیم برگ و سوخ، با افزایش تنشها به ویژه تنش شوری، افزایش یافت.نتیجهگیری: نتایج نشان داد که همه سطوح تنشهای خشکی و شوری موجب افزایش و بهبود فعالیت آنزیمهای آنتیاکسیدانی (کاتالاز و گایاکول پراکسیداز) و غیر آنزیمی (فلاونوئید کل) گل نرگس شد؛ اما تحت شرایط تنش خشکی و شوری، قطر گل، میزان کاروتنوئید و کلروفیل کل کاهش یافتند. تحت شرایط تنش شوری و خشکی، میزان تجمع سدیم در برگ در مقایسه با سوخ، بیشتر بود. نتایج نشان داد حساسیت گیاه نرگس نسبت به تنش شوری بیشتر از خشکی بود که با کاربرد همزمان دو تنش، اثرات منفی تشدید شد. بهطور کلی، نتایج نشان داد که کشت گل نرگس تا سطح رطوبتی 70 درصد ظرفیت زراعی و شوری آب آبیاری حدود 3 دسی زیمنس بر متر تأثیر منفی قابل توجهی بر عملکرد و کیفیت گیاه نداشت و قابل توصیه است.

اثر تلقیح باکتری Rhizobium leguminosarum در ترکیب با سطوح مختلف جمعیت باکتری Peseudomonas fluorescens بر برخی صفات کمی باقلا (Vicia faba)
چکیدهسابقه و هدف: استفاده از باکتریها بهعنوان کود زیستی یکی از مسائل مهم در توسعه کشاورزی پایدار محسوب میشود که مهمترین نتیجه آن کاهش آلودگی محیط زیست در اثر کاهش مصرف کودهای شیمیایی است. گیاه باقلا سالهاست به دو دلیل بسیار مهم، ارزش تغذیهای دانهها و ارزش اکولوژیک آن جهت تثبیت نیتروژن، در تناوبهای زراعی با غلات مورد استفاده قرار میگیرد. تحقیق حاضر با هدف بررسی اثر سطوح مختلف جمعیت باکتری Peseudomonas fluorescens به تنهایی و در ترکیب با باکتری Rhizobium leguminosarum بر برخی فاکتورهای رشد و عملکرد باقلا، جهت حصول عملکرد مناسب اجرا گردید.مواد و روشها: آزمایش طی سالهای 95-1394 و 96-1395 در قالب طرح پایه بلوک کامل تصادفی و به صورت کرتهای خردشده، در مزرعهای واقع در ایستگاه تحقیقات کشاورزی استان گلستان، اجرا شد. Rhizobium leguminosarum به عنوان کرت اصلی و مقادیر باکتری Peseudomonas fluorescens در پنج سطح صفر، 103×9، 105×9، 107×9، 109×9 باکتری زنده در هر میلیلیتر مایه تلقیح، بهعنوان کرت فرعی در نظر گرفته شد. صفت ارتفاع بوته در زمان رسیدگی فیزیولوژیک، شاخص سطح برگ در زمان غلاف دهی، تعداد دانه و غلاف در مترمربع در یک نمونه شامل 10 بوته اندازهگیری و وزن 100 دانه نیز در یک نمونه 100 تایی توزین گردید. عملکرد دانه با احتساب رطوبت 11 درصد سال اول و 9 درصد سال دوم، عملکرد غلاف سبز و عملکرد زیستی در واحد سطح بدست آمد. از نسبت عملکرد اقتصادی(دانه) به عملکرد زیستی، شاخص برداشت محاسبه گردید.یافتهها: نتایج دوسال نشان داد، اثر باکتریها، تحت تأثیر شرایط اقلیمی قرار گرفتهاست. در سال اول، تعداد غلاف و دانه در مترمربع تحت تأثیر اثر متقابل ریزوبیوم و سودوموناس قرار گرفتند و بهترین تیمار، کاربرد ریزوبیوم بههمراه تعداد 105×9، باکتری زنده Peseudomonas fluorescens در هر میلیلیتر مایه تلقیح بود. اثر سطوح مختلف جمعیت باکتری سودوموناس روی عملکرد غلاف سبز، در سطح احتمال 5 درصد معنیدار شد. با کاربرد تعداد 105×9 باکتری سودوموناس، افزایش 14 درصدی در عملکرد غلاف سبز نسبت به شاهد مشاهده شد. با افزایش تعداد باکتری (107×9 باکتری زنده سودوموناس در میلی-لیتر مایه تلقیح) کاهش معنیداری در عملکرد غلاف نسبت به تیمار 105×9، دیده شد و در نهایت در بالاترین تعداد (109×9 باکتری زنده در میلیلیتر مایه تلقیح)، سودوموناس باعث افزایش دوباره و معنیدار عملکرد مذکورگردید که با تیمار 105×9 باکتری زنده در میلیلیتر مایه تلقیح، تفاوت معنیداری نداشت. در سال اول، کاربرد ریزوبیوم باعث افزایش 10 درصدی عملکرد زیستی نسبت به شاهد گردید که در سطح احتمال 5 درصد معنیدار شد. نتایج سال دوم نشان داد که از کارایی ریزوبیوم کاسته شده و بیشترین تعداد دانه در متر مربع، عملکرد زیستی و عملکرد دانه در واحد سطح، با کاربرد تعداد 105×9 باکتری زنده سودوموناس در هر میلیلیتر مایه تلقیح بدست آمد که با جمعیتهای بالاتر بهلحاظ آماری تفاوتی نداشت. تعداد غلاف در مترمربع نیز تحت تأثیر معنیدار و متقابل هر دو باکتری ریزوبیوم و سودوموناس قرار گرفت. نتیجهگیری: این مطالعه نشان داد که در مجموع اثر Peseudomonas fluorescens بر عملکرد و اجزای آن در شرایط محیطی مختلف دو سال، از ثبات بیشتری نسبت به ریزوبیوم برخوردار بود و بهترین جمعیت باکتریPeseudomonas fluorescens جهت تلقیح بذور باقلا، 105 × 9 باکتری زنده در هر میلیلیتر مایه تلقیح، مشخص شد که بهنظر میرسد آستانه پاسخ باقلا به این باکتری باشد.

ارزیابی صفات کمی و کیفی در برخی از ژنوتیپ های توت فرنگی با راهکار PLS-PM
سابقه و هدف: تعیین ارتباط بین ویژگی های مورفولوژیکی، فیزیولوژیکی و بیوشیمیایی (کیفی)، جهت حصول افزایش تولید و کیفیت محصولات باغی از جمله توت فرنگی ضرورت دارد. با گذشت زمان، راهکارهای آماری متنوعی جهت درک روابط واقعی متغیرها ظهور پیدا کرده و راهکارهای آماری قدیمی تر، بتدریج جای خود را به انواع جدید، به دلیل برآورد و حصول علمی تر و مطمئن تر نتایج داده اند. در این بین، راهکار جدید PLS-PM بدلیل روش نوین و نتایج معتبر و جذاب تر، کاربرد زیادی نسبت به روش های آماری قدیمی دارد. مواد و روش ها: صفات مورفولوژیکی: گل، میوه، کیفی (بیوشیمیایی) و عملکرد در 8 ژنوتیپ توتفرنگی طی 4 سال در قالب طرح بلوکهای کامل تصادفی با سه تکرار مطالعه گردید. از راهکارهای مختلف آماری همانند برآورد ضرایب همبستگی، رگرسیون گامبهگام، تجزیه مسیر ساده و جهت درک دقیقتر روابط، الگوریتم تحلیل داده ها با راهکار پیشرفته ی PLS-PM (شامل آلفای کرونباخ، ضرایب بار عاملی، معناداری ضرایب مسیر و ضریب تبیین، نیکویی برازش و بارهای عاملی متقاطع) استفاده گردید. یافته ها: با وجود تنوع ژنتیکی بین ارقام، بین عملکرد میوه با ویژگی های میوه، تعداد برگ، ویژگی های گل آذین و تعداد طوقه در بوته و نیز بین اندازه میوه و میزان آنتوسیانین همبستگی بالا و مثبتی مشاهده شد. تعداد گل آذین، اندازه و حجم و تعداد میوه، تاریخ تشکیل ساقهرونده و طول دوره گلدهی 3/88 درصد از تغییرات عملکرد را تبیین و مهم ترین اجزاء آن شناخته شدند. تعداد وحجم میوه بیشترین اثر مستقیم را بر افزایش عملکرد میوه داشتند. گل دهی، بیشترین تأثیر را بر عملکرد میوه داشته و سپس صفات موروفولوژیکی بیشترین تأثیر را بر صفات بیوشیمیایی و گلآذین داشتند. اندازه و حجم میوه، بیش از بقیه، تحت تأثیر صفات مورفولوژیکی بود. نتیجه گیری: در این تحقیق، از راهکارهای آماری مختلفی جهت درک ارتباطات صفات با ماهیت متفاوت توت فرنگی استفاده گردید، که هیچکدام به اندازه راهکار جدید ولی ناشناخته PLS-PM نشان دهنده ی تأثیر مهم صفات گل دهی در افزایش عملکرد میوه توت فرنگی نبود. با بکارگیری روش جدید PLS-PM در مقایسه با روشهای قدیمی آماری، و با انجام دستهبندی صفات، با توجه به ماهیت متفاوت آنها، نتیجه گرفته شد که در بحث افزایش عملکرد میوه توت فرنگی، صفات گل دهی از بقیه صفات موردنظر، مؤثرتر بوده و صفات ریختی شامل: ارتفاع گلآذین، تعداد برگ، طول گوشوارک، تعداد طوقه، تاریخ شروع و تعداد ساقهرونده و سطح برگ نیز از جمله صفات تأثیرگذار بوده که بر اساس آنها میتوان برنامههای اصلاحی و تولید اقتصادی توتفرنگی را مدیریت نمود.

بررسی تاثیر غلظتهای کاهش یافته گلایفوسیت و سولفوسولفورون بر کنترل گلجالیز مصری (Phelipanche aegyptiaca Pers.) در خیار (Cucumis sativus L.)
سابقه و هدف: گلجالیز گیاهی فاقد کلروفیل و انگل مطلق ریشه گیاهان دولپهای است که دامنه پراکندگی آن بیشتر در نواحی گرم و خشک و همچنین نواحی معتدل و نیمه خشک کشورهای مدیترانهای، اروپای شرقی و جنوبی و خاورمیانه از جمله ایران آلودگی ایجاد میکند. علفهای هرز انگلی به ویژه گلجالیز به دلیل کاهش عملکرد و افزایش هزینه در تولید خیار، از اهمیت ویژهای برخوردار است. هدف از انجام این مطالعه کنترل شیمیایی گل جالیز با علفکشهای گلایفوسیت و سولفوسولفورون و تعیین میزان مناسب و تعداد دفعات کاربرد بوده. بر همین اساس آزمایشی در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه کردستان اجرا شد.مواد و روشها: این آزمایش در بهار و تابستان (25 اردیبهشت تا پایان شهریور) سال 1397 به صورت گلدانی در فضای آزاد مزرعه، در قالب طرح بلوکهای کامل تصادفی و با چهار تکرار که هر تکرار شامل 25 تیمار علفکشی به شرح زیر بود: تیمار 1) شاهد آلوده به گلجالیز و بدون اعمال علفکش، تیمارهای 2 تا 5) علفکش گلایفوسیت (SL 36%) 20، 40، 60 و 80 گرم ماده موثره در هکتار یکبار کاربرد، تیمارهای 6 تا 9) علفکش سولفوسولفورون (WG 75%) 25، 50، 75 و 100 گرم ماده موثره در هکتار یکبار کاربرد، تیمارهای 10 و 11) علفکش گلایفوسیت 20 و 40 گرم ماده موثره در هکتار دوبار کاربرد، تیمارهای 12 و 13) علفکش سولفوسولفورون 25 و50 گرم ماده موثره در هکتار دوبار کاربرد، تیمارهای 14 تا 16) علفکش گلایفوسیت 20، 40 و 60 گرم ماده موثره در هکتار سهبار کاربرد، تیمارهای 17 تا 19) علفکش سولفوسولفورون 25، 50 و 75 گرم ماده موثره در هکتار سهبار کاربرد، تیمارهای ترکیبی20 تا 22) علفکشهای سولفوسولفورون 25 + گلایفوسیت 20 گرم ماده موثره در هکتار دوبار کاربرد، گلایفوسیت 40 + سولفوسولفورون 50 گرم ماده موثره در هکتار دوبار کاربرد و سولفوسولفورون 50 + گلایفوسیت 40 گرم ماده موثره در هکتار دوبار کاربرد و تیمارهای ترکیبی 23 تا 25) علفکشهای گلایفوسیت 20 + سولفوسولفورون 25 + گلایفوسیت 40 گرم ماده موثره در هکتار سهبار کاربرد، سولفوسولفورون 25 + گلایفوسیت 40 + سولفوسولفورون 50 گرم ماده موثره در هکتار سهبار کاربرد و گلایفوسیت 40 + سولفوسولفورون 50 + گلایفوسیت 40 گرم ماده موثره در هکتار سهبار کاربرد. اولین مرحله اعمال تیمارها 15 روز بعد از سبز شدن بذرهای خیار بود و فاصله بین دو تکرار سمپاشی 14 روز بود. یافتهها: نتایج این مطالعه نشان داد که خیار به علفکش سولفوسولفورون حساسیت بیشتری داشته است. کاربرد سولفوسولفورون 75 گرم ماده موثره در هکتار یکبار سمپاشی، سولفوسولفورون 25، 50 و 75 گرم ماده موثره در هکتار سهبار سمپاشی ، سولفوسولفورون 50 + گلایفوسیت 40 گرم ماده موثره در هکتار دوبار سمپاشی، وتیمارهای ترکیبی سهبار سمپاشی در تمام صفات مورد اندازهگیری در میزبان و گلجالیز نسبت به تیمار شاهد در سطح یک درصد معنیدار بوده با وجود کنترل گلجالیز گیاهسوزی زیادی بر روی گیاه میزبان بر جای گذاشتند و به همین دلیل استفاده از آنها توصیه نمیشود. به دلیل حساس بودن خیار و اثر سمیت کمتر تیمارهای گلایفوسیت 20 گرم ماده موثره در هکتار یک و سهبار کاربرد و گلایفوسیت 40 گرم ماده موثره در هکتار یکبار کاربرد به ترتیب با 247، 215 و 151 درصد افزایش نسبت به تیمار شاهد عملکرد بهتری از خود نشان دادند و علاوه بر کنترل گلجالیز، گیاهسوزی کمتری هم بر روی گیاه میزبان مشاهده شد.نتیجهگیری: بطورکل بهترین نتایج با در نظر گرفتن کنترل گلجالیز مصری و عدم خسارت به گیاه میزبان، کاربرد مقادیر گلایفوسیت 20 گرم ماده موثره در هکتار یک و سهبار کاربرد و گلایفوسیت 40 گرم ماده موثره در هکتار یکبار کاربرد به دست آمد.

کلونینگ و توالی یابی ژن p- انسولین جدا شده از گیاه دارویی Momordica charantia و تحلیل بیوانفورماتیکی ساختار سه بعدی پروتئین
چکیده سابقه و هدف:دیابت بیماری است که بدن توانایی کنترل قند خون را از دست می دهد. داروهای شیمیایی تجویز شده به منظور درمان قند خون دارای اثرات جانبی نامطلوب متعددی هستند. در این زمینه داروهای گیاهی جایگزین بسیار مناسبی میباشند چون اثرات جانبی بسیار کم تری دارند. گیاه کارلا با نام علمی Momordica charantia از خانواده Cucurbitaceae می باشد. بذرهای کارلا دارای پلی پپتیدی به نام p-انسولین می باشد که از 172 اسید آمینه تشکیل یافته است. مقدار p-انسولین بسته به بافت گیاه، رقم، محل کشت و فصل برداشت بسیار متغیر است و در مجموع مقدار آن نسبتا پایین میباشد بطوریکه این میزان برای تولید صنعتی داروها بر پایه این ترکیب کافی نیست. علاوه بر این به دلیل وجود ترکیبات مداخله گر مانند پلی ساکارید ها، ممکن است این ماده موثره تاثیر کمتری داشته باشد. کلون کردن ژن p- انسولین و بیان هترولوگ آن در میکروارگانیسم ها، راه حلی سریع و مفید برای حل این مشکل است. بدین منظور در این تحقیق توالی ژن p- انسولین از گیاه کارلا از طریق RT-PCR شناسایی و سپس توالی یابی شد.مواد و روش ها: به منظوراستخراج RNA ، در دو مرحله نارس و رسیده از فرابر، آریل بذر و بذر گیاه کارلا نمونه گیری انجام شد. کمیت و کیفیت RNA استخراج شده از طریق دستگاه اسپکتروفوتومتری و الکتروفوز بر روی ژل TBE 1% تعیین شد. سپس جهت کلونینگ ژن p-انسولین، RT-PCR با پرایمرهای اختصاصی ژن p-انسولین انجام شد. سپس محصول RT-PCR ژن p-انسولین ، از طریق ژل الکتروفورز خالص سازی و کلونینگ ژن p-انسولین در تی وکتور صورت گرفت. پس از استخراج پلاسمید نوترکیب و تایید حضور ژن p-انسولین در تی وکتور، پلاسمید نوترکیب تعیین توالی شد. در ادامه ساختار سه بعدی پروتئین p-انسولین از طریق مدلینگ به کمک نرم افزار Phyre 2.0 (www.sbg.bio.ic.ac.uk/phyre2/) بررسی شد.یافته ها:در نتایج بدست آمده ازالکتروفورز RNA از بافت های مختلف کارلا، وجود دو باند مربوط به 28SrRNA و 18SrRNA نشان دهنده استخراج موفق RNA بود. نتایج RT-PCR با پرایمرهای اختصاصی ژن p-انسولین باند اختصاصی bp750 موید تکثیر صحیح cDNA بود. همچنین، بیان ژن p-انسولین در بافت آریل بذر رسیده بیشتر از سایر بافت های میوه بود. پس از خالص سازی باند مربوط به ژن p-انسولین ، واکنش لیگاسیون ژن هدف در داخل تی وکتور و ترانسفورماسیون آن به داخل میزبان E. coli صورت گرفت که نتایج مثبت موید انجام موفق کلونینگ بود. در ادامه تعیین توالی ژن p-انسولین، از گیاه دارویی کارلا برای اولین بار در ایران انجام شد. نتایج بیوانفوماتیک نشان داد که دومین های فعال منواکسیژناز متصل شونده به FDA در بخش N ترمینال و پرولیپوپروتئین دی آسیل گلیسریل ترانسفراز در C ترمینال پروتئین p- انسولین وجود دارد. نتیجه گیری:بیان کم پروتئین p- انسولین در گیاه کارلا و وجود ترکیبات مداخله کننده مانند پلی ساکاریدها ممکن است باعث کاهش تاثیر ماده موثره کاهنده قند خون می شود. لذا، در این تحقیق ژن p- انسولین از گیاه کارلا کلون و تعیین توالی شد. تعیین توالی ژن p- انسولین امکان بیان هترولوگ آن در میکروارگانیسم و تولید فراوان و خالص پروتئین p- انسولین را به عنوان دارو فراهم خواهد کرد.واژه های کلیدی: Momordica charantia، دیابت، کلونینگ، توالی یابی، بیوانفورماتیک
