The model plant Arabidopsis thaliana contains seven RecQ homologues. We already showed that inactivation of AtRecQl4A alters plant responses to genotoxic stress. In this report, some physiological and biochemical parameters in recQl4A mutant plants were investigated under salinity treatment. Wild type and recQl4A mutant plants were grown on Murashige and Skoog agar plates containing various concentrations of NaCl arranged in a completely randomized design. When plants were treated with 100-150 mM NaCl, damage to the plants was so high that we were not able to measure physiological or biochemical parameters accurately. In 75 mM NaCl, total soluble protein in all examined plant lines declined. In response to salinity treatment, mutant seedlings exhibited less dry weight and chlorophyll content, as compared to the wild type plants. However, total soluble sugar in the mutant seedlings was higher than that in wild type plants. Similarly, the catalase activity in the mutant plants was significantly higher than that of wild type, when plants were treated with 75 mM NaCl. Interestingly, increasing of NaCl concentration form 0 to 75 mM caused a significant reduction in the polyphenol oxidase activity in the mutant plants but not in the wild type plants. These results propose that the RecQl4A gene plays crucial roles in response of Arabidopsis plants to NaCl treatment and provide new insights in understanding mechanisms underlying plant salinity tolerance.}, keywords = {Arabidopsis,RecQl4A,catalase,Peroxidase,Polyphenoloxidase}, title_fa = {بررسی برخی از پارامترهای فیزیولوژیک و بیوشیمیایی در گیاهان جهش‌یافته AtrecQl4A تحت تنش شوری}, abstract_fa = {اعضای خانواده RecQ در ترمیم، رونویسی و نوترکیبی DNA وظایف مهمی بر عهده دارند. گیاه مدل ارابیدوپسیس دارای 7 همولوگ RecQ است. ما قبلاً نشان دادیم که از کار افتادن ژنRecQl4A اربیدوپسیس پاسخ گیاه به برخی از تنش‌های ژنوتوکسیک را تغییر می‌دهد. در این گزارش، برخی از پارامترهای فیزیولوژیک و بیوشیمیایی در گیاه جهش‌یافتهrecQl4A  تحت تیمار شوری اندازه‌گیری شد تا بتوان نقش ژن RecQl4A در کنترل پاسخ گیاه به استرس شوری را تعیین نمود. گیاهان وحشی و جهش‌یافته در قالب یک طرح کاملاً تصادفی روی پلیت‌های آگار- موراشی- اسکوگ حاوی NaCl رشد داده شدند. هنگامی‌که گیاهان با 100 و 150 میلی‌مولار NaCl تیمار شدند صدمات وارده به گیاهان به قدری زیاد بود که امکان اندازه‌گیری پارامترهای فیزیولوژیک و بیوشیمیایی وجود نداشت. در غلظت 75 میلی‌مولار کلرید سدیم، مقدار پروتئین کل محلول در پاسخ به شوری کاهش یافت. به‌علاوه، در پاسخ به شوری، گیاهان جهش‌یافته، وزن خشک و محتوای کلروفیل کمتری در مقایسه با گیاهان وحشی داشتند؛ لیکن قند کل در گیاهچه‌های جهش‌یافته از گیاهچه‌های وحشی بیشتر بود. به همین ترتیب هنگامی‌که گیاهان با 75 میلی‌مولار کلرید سدیم تیمار شدند میزان فعالیت آنزیم کاتالاز در گیاهان جهش‌یافته از فعالیت آن در گیاهان وحشی بیشتر بود. جالب توجه این‌که افزایش غلظت NaCl از صفر به 75 میلی‌مولار باعث شد تا فعالیت آنزیم پلی‌فنل‌اکسیداز فقط در گیاهان جهش‌یافته به‌طور معنی‌داری کاهش یابد. این نتایج پیشنهاد می‌کند ژنRecQl4A  در تنظیم پاسخ گیاه آرابیدپسیس به تیمارNaCl  نقش اساسی ایفا می‌کند و بدین‌ترتیب دانش محققان را در درک سازوکار‌های کنترل‌کننده مقاومت گیاهان به تنش‌های محیطی افزایش می‌دهد.}, keywords_fa = {آرابیدوپسیس,RecQl4A,کاتالاز,پراکسیداز,پلی‌فنل‌اکسیداز}, url = {https://jopp.gau.ac.ir/article_334.html}, eprint = {https://jopp.gau.ac.ir/article_334_52800c2bd6a7e03d95ff581c2a4daad0.pdf} }
