مطالعه تاریخ سوخدهی و صفات مرفولوژیک جمعیتهای بومی و ارقام تجاری پیاز در خوزستان
سابقه و هدف: پیاز با سطح زیر کشت 63685 هکنار یکی از مهمترین سبزیها در ایران میباشد. بخش قابل توجهی از بذر ارقام روز کوتاه پیاز از طریق واردات تامین میشود که شرایط را برای حذف و فرسایش ژنتیکی تودههای بومی فراهم نموده است. اما علی-رغم این فرسایش، تنوع وسیعی در این تودهها گزارش شده است. در بین این تودهها، تودههایی با عملکرد بالا، خاصیت انبارمانی طولانی و مقاوم به آفات در مطالعات گوناگون مشاهده شده است، بنابراین مطالعه کمی و کیفی این تودهها حائز اهمیت فراوانی می-باشد. در همین راستا این پژوهش به منظور مطالعه تاریخ سوخدهی، صفات مرفولوژیک و همبستگی صفات جمعیتهای بومی جنوب کشور و دو رقم تجاری پریماورا و تگزاسارلیگرانو صورت گرفت. مواد و روشها: این تحقیق بهصورت طرح بلوکهای کامل تصادفی شامل هشت تیمار (پیاز اصلاح شده بهبهان، تودههای محلی بهبهان، پادوک، رامهرمز، برازجان، ایرانشهر و ارقام تگزاسارلیگرانو و پریماورا) با چهار تکرار در ایستگاه تحقیقات کشاورزی بهبهان بهمدت دو سال زراعی (93-1392) اجرا شد. بذور در اواسط مهر ماه در خزانه کشت و نشاها (در مرحله 2 تا 3 برگی) و در اواخر آذر ماه به زمین اصلی منتقل شدند. تاریخ تشکیل سوخ با شاخص نسبت تشکیل سوخ و مجموع تجمعی تخمین زده شد. سوخها در زمان افتادگی50 تا 80 درصد برگها و شروع خشک شدن آنها برداشت شدند. نتایج حاصله توسط نرم افزار MSTATC تجزیه و تحلیل گردید و میانگینها با استفاده از آزمون چند دامنهای دانکن مقایسه شدند.یافتهها: در سال اول آزمایش در جمعیتهای مورد مطالعه سوخ از 27 اسفند با طول روز 11 ساعت و 56 دقیقه تا 3 فروردین با طول روز 12 ساعت و 5 دقیقه تشکیل شد. در سال دوم آزمایش زمان سوخدهی از 12 فروردین با طول روز 12 ساعت و 32 دقیقه تا 30 فروردین با طول روز 13 ساعت و 5 دقیقه متغیر بود. حداکثر عملکرد کل در سال اول و دوم آزمایش بهترتیب توسط توده محلی پادوک و رقم تگزاسارلیگرانو تولید شد. بیشترین عملکرد قابل فروش به رقم پریماورا مربوط بود. در ارقام تگزاسارلیگرانو و پریماورا بولتینگ مشاهده نگردید. حداکثر و حداقل دوقلویی بهترتیب به رقم پریماورا و توده محلی برازجان مربوط بود. بیشترین درصد ماده خشک سوخ به پیاز اصلاح شده بهبهان تعلق داشت. مطالعه ضرایب همبستگی مشخص نمود عملکرد کل و قابلفروش با بولتبنگ و دوقلویی همبستگی منقی و معنیداری دارند. بین درصد ماده خشک سوخ و درصد کل مواد جامد محلول سوخ رابطه مثبت و معنی-داری مشاهده گردید. نتیجهگیری: همه جمعیتهای مورد مطالعه روز کوتاه بودند. بر اساس نتایج این بررسی برای کشت پیاز در خوزستان رقم تگزاسارلی گرانو بهترین رقم میباشد. در میان جمعیتهای بومی، پیاز اصلاح شده بهبهان، از نظر حساسیت پایین به دوقلویی و بالا بودن درصد ماده خشک سوخ، مناسبترین جمعیت میباشد.

بررسی راهکارهای زراعی تراکم بوته و سطح کود نیتروژن بر تولید کلزای پاییزه (Brassica napus L.) در شرایط تداخل با جمعیت علف هرز ارشته خطایی (Lepyrodiclis holosteoides Fenzl.)
سابقه و هدف: مدیریت کلزا برای تولید عملکرد بالای دانه همراه با کیفیت مطلوب، مستلزم توجه دقیق به ارزیابی وضعیت تغذیهای گیاه به خصوص مقدار نیتروژن طی فصل رشد، تعیین تراکم مطلوب و کنترل علفهای هرز میباشد. یکی دیگر از عوامل مهم مدیریت زراعی جهت دستیابی به عملکرد بالا در کلزا، کنترل علفهای هرز میباشد. عدم وجود علفکشهای اختصاصی برای کنترل علفهای هرز در این محصول سبب خسارت هنگفت به این محصول کلزا شده است. علف هرز ارشته خطایی به عنوان یکی از علفهای هرز مهاجم در چند سال اخیر در مزارع کلزا شایع شده است. با وجود گسترش سریع این علف هرز در کشور تاکنون پژوهشی در ارتباط با رقابت آن با کلزا نشده است. لذا این آزمایش با هدف بررسی اثرات مقادیر نیتروژن و تراکم کلزا در شرایط رقابت با علف هرز ارشته خطایی انجام شد.مواد و روش: این آزمایش در سال زراعی 95-1394 در مزرعه پژوهشی مرکز آموزش عالی امام خمینی کرج انجام شد. آزمایش به صورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی با سه تکرار اجرا شد. تیمارها شامل مقدار نیتروژن از منبع کودی اوره (46 درصد نیتروژن) در چهار سطح صفر، 50، 100 و 150 کیلوگرم نیتروژن خالص در هکتار، تراکم کلزا در دو سطح 70 و 90 بوته در متر مربع و تراکم علف هرز ارشته خطایی در چهار سطح صفر، 5، 10 و 15 بوته در متر مربع بود.یافتهها: مقایسه میانگین اثر متقابل تراکم کلزا و سطوح نیتروژن نشان داد که بیشترین تعداد خورجین در بوته، تعداد دانه در خورجین و عملکرد دانه، در تراکم کلزا 90 بوته در متر مربع با مصرف 100 و 150 کیلوگرم نیتروژن در هکتار بدست آمد. همچنین نتایج نشان داد که در تراکمهای 5، 10 و 15 بوته در متر مربع علف هرز ارشته خطایی، بهترتیب 11، 19 و 32 درصد در تراکم کلزای 70 بوته و بهترتیب 5، 9 و 14 درصد در تراکم کلزای 90 بوته در متر مربع سبب کاهش عملکرد دانه کلزا نسبت به تیمار عدم حضور علف هرز شد. نتایج اثر سطوح نیتروژن و تراکم علف هرز ارشته خطایی نشان داد که بیشترین عملکرد دانه با 3335 کیلوگرم در هکتار در تیمار 150 کیلوگرم نیتروژن در هکتار و بدون حضور ارشته خطایی بدست آمد که با حضور 5، 10 و 15 بوته در متر مربع از این علف هرز عملکرد دانه بهترتیب 8، 14 و 28 درصد کاهش نشان داد. نتیجهگیری: بطورکلی نتایج نشان داد که در هر دو تراکم کلزا با افزایش کود نیتروژن تا 100 کیلوگرم در هکتار علاوه بر بهبود عملکرد دانه و اجزای عملکرد سبب افزایش توان رقابتی کلزا در مقابل علف هرز مهاجم ارشته خطایی خواهد شد.

تأثیر تنظیمکنندههای رشد بر ریزازدیادی پایهرویشی GF677 در شرایط محیطکشت مایع
ممقدمه: پایه GF677 (دورگ بین هلو و بادام)، یک پایه مناسب برای هلو، شلیل و بادام است که به طور گسترده در سراسر دنیا استفاده می-شود. به دلیل کارایی کم ازدیاد از طریق قلمه، کشتبافت روشی مناسب و سریع برای ازدیاد این پایه است. در محیط مایع تماس بافت گیاهی با محیطکشت بهتر، تکثیر سریعتر، و هزینه تولید کاهش مییابد. بنابراین هدف این تحقیق بهبود پرتکل ریزازدیادی پایه GF677، با استفاده از محیطکشت مایع بود.مواد و روشها: در این تحقیق ریزنمونههای گره ضدعفونی شده در محیطکشتهای MS، B5 و WPM در حالت مایع حاوی هورمون-های BA (بنزیلآدنین)، در پنج غلظت 25/0، 5/0، 1، 2 و 4 میلیگرم بر لیتر و IBA (ایندول-3- بوتیریک اسید)، در چهار غلظت 0، 1/0، 25/0 و 5/0 میلیگرم بر لیتر بررسی شد. آنالیز دادهها اساس روش آماری فاکتوریل در قالب طرح کاملاٌ تصادفی و مقایسات میانگین دادهها به روش آزمون LSD در سطح احتمــال 5% انجام شد. نتایج و بحث: نتایج نشان داد که حداکثر تعداد شاخساره (44/2 شاخساره) در محیط مایع WPM با غلظت 1 میلیگرم در لیتر BA و بلندترین طول شاخسارهها در محیطکشت حاوی 1 میلیگرم در لیتر BA و 25/0 میلیگرم در لیتر IBA در محیطکشت B5 مشاهده شد. این مطالعه نشان داد تعداد شاخساره با افزایش غلظت BA تا غلظت مشخصی افزایش یافت. به نظر میرسد که بین غلظت BA و تعداد شاخساره تا یک غلظت خاصی از BA، رابطه مثبت وجود دارد به طوری که تعداد شاخساره در غلظت 1 میلیگرم در لیتر BA به بیشترین تعداد خود رسید. در غلظتهای بالاتر از 1 میلیگرم در لیتر BAP، تعداد شاخسارهها کاهش یافت. به نظر میرسد پایه GF 677 دارای مقادیری از تنظیمکنندههای رشد به صورت درونزا باشند، به طوری که کاربرد غلظت کمی از تنظیم کنندههای رشد برونزا برای پرآوری آنها کافی است . به نظر می_رسد مقدار مشخصی از BA برای به دست آوردن بیشترین باززایی مورد نیاز است و غلظتهای بالاتر BA سبب ایجاد مقدار زیادی کالوس میشود که در کشتبافت برای تولید شاخساره مناسب نیست.جهت ریشهزایی، شاخسارهها را به محیطکشت MS جامد حاوی 1 میلیگرم در لیتر IBA و 5/0 میلیگرم در لیتر BA منتقل و 33 درصد ریشهزایی حاصل شد. گیاهچههای ریشهدار شده در محیط شامل پرلیت، ماسه و خاک به نسبت 1:2:1 قرار گرفتند. 90 درصد نمونههای انتقال یافته به محیط خاکی زنده مانده و رشد طبیعی داشتند. نتیجهگیری: ریزازدیادی پایه GF677 تحت تأثیر محیطکشت (MS، B5 و WPM) و تنظیمکنندههای رشد قرار گرفت. محیطکشت WPM همراه با 1 میلیگرم در لیتر BA با صددرصد باززایی ریزنمونهها و تولید بیشترین تعداد شاخساره (44/2 شاخساره)، مناسبترین محیط-کشت در بین سایر محیطهای کشت مورد استفاده بود.

اثر نوع ریزنمونه و تنظیم کنندههای رشد بر کالوسزایی و متابولیتهای ثانویه کاسنی (Cichorium intybus L.)
سابقه و هدف: گیاه کاسنی (Cichorium intybus L.) یک گیاه دارویی مهم دارای متابولیتهای با ارزش است. کالوس کاسنی میتواند منبع خوبی برای تولید و استخراج متابولیتهای ثانویهی این گیاه باشد و غلظت تنظیم کنندههای رشد و نوع ریزنمونه از عوامل اصلی اثرگذار در تولید کالوس کاسنی میباشند. هدف این پژوهش، بهینهسازی کالوسزایی و تولید متابولیتهای ثانویه در گیاه کاسنی با استفاده از غلظت تنظیم کنندههای رشد و نوع ریزنمونه است.مواد و روشها: اثر غلظت تنظیم کنندههای رشد بنزیل آدنین (BA) (۰، ۵/۰، ۱ و ۲ میلیگرم در لیتر) و نفتالین استیک اسید (NAA) (۰، ۵/۰ و ۱ میلیگرم در لیتر) و نوع ریزنمونه (دمبرگ، ساقه، برگ) بر کالوسزایی و میزان تولید فنل و فلاونوئید کاسنی در آزمایشی به صورت فاکتوریل و در قالب طرح کاملاً تصادفی با سه تکرار مورد بررسی قرار گرفت. نخست سطح بذرها سترون گردید و سپس در محیط MS به منظور تهیهی گیاهچهی سترون کشت شدند. از این گیاهچهها، پس از 4 تا 5 هفته، ریزنمونههای برگ، دمبرگ و ساقه در شرایط سترون تهیه شد. ریزنمونهها در محیط کشت بافت گیاهی حاوی غلظتهای مختلفBA و NAA کشت شدند. پس از 4 هفته، کالوسهای بدست آمده از ریزنمونههای استریل برای اندازهگیری صفات درصد کالوسزایی، وزن تر، وزن خشک، درصد ماده خشک، محتوای آب نسبی کالوس، میزان فنل، فلاونوئید، فعالیت آنتیاکسیدانی مورد استفاده قرار گرفتند.یافتهها: بر اساس نتایج به دست آمده از تجزیه واریانس، اثر متقابل همه تیمارها اختلاف معنی داری را در سطح یک درصد نشان دادند. مقایسه میانگینهای اثر متقابل تیمارها نشان داد که ریزنمونهی دمبرگ در تیمار ۲ میلیگرم در لیتر BA همراه با ۱ میلیگرم در لیتر NAA بیشترین میزان کالوسزایی را داشت. از طرف دیگر، بیشترین وزن تر کالوس در ریزنمونهی برگ در محیط کشت حاوی ۵/۰ میلیگرم در لیتر BA و ۵/۰ میلیگرم در لیتر NAA بدست آمد. همچنین بیشترین وزن خشک کالوس در ریزنمونهی دمبرگ و ساقه در غلظتهای ۱ و ۲ میلیگرم در لیتر BA و ۵/۰ میلیگرم در لیتر NAA بدست آمد. بیشترین درصد ماده خشک کالوس در ریزنمونهی دمبرگ در غلظت۱ میلیگرم در لیترBA و ۵/۰ میلیگرم در لیتر NAA مشاهده شد. بیشترین میزان فنل و فلاونوئید در ریزنمونهی دمبرگ و غلظت ۵/۰ میلیگرم در لیتر BA به تنهایی مشاهده گردید. کالوس تولید شده از ریزنمونهی ساقه بیشترین خاصیت آنتی اکسیدانی را در محیط کشت حاوی ۱ میلیگرم در لیترBA و ۱ میلیگرم در لیتر NAA، نشان داد. نتیجه گیری کلی: در مجموع، دمبرگ بهترین ریزنمونه برای تولید کالوس در ترکیب با تیمارهای ۲ میلیگرم در لیتر BA و ۱ میلیگرم در لیتر NAA و یا محیط حاوی ۵/۰ میلیگرم در لیتر BA و بدون NAA بود. افزون بر این، تیمارهای فوق، بیشترین میزان فنل و فلاونوئید را تولید نمودند که دارای بیشترین فعالیت آنتی اکسیدانی هم بودند.

بررسی تنوع ژنتیکی برخی ژنوتیپهای مرکبات ایران بر اساس خصوصیات مورفولوژی و نشانگرهای مولکولی ISSR و PCR-RFLP
سابقه و هدف: ژرمپلاسم مرکبات ایران تنوع ژنتیکی گستردهای دارد که ناشی از دگرگردهافشانی، سابقه طولانی ازدیاد بذری و فراوانی جهشهای ژنتیکی است. برای تعیین وضعیت ردهبندی، روابط فیلوژنتیک و فاصله ژنتیکی بین افراد این ذخیره ارزشمند ژنتیکی باید از ویژگیهای مورفولوژی در کنار نشانگرهای مولکولی مبتنی بر DNA استفاده کرد. در تحقیق حاضر برای کسب اطلاعاتی در مورد درجه قرابت ژنتیکی موجود بین 79 ژنوتیپ ناشناخته محلی مرکبات موجود در کلکسیون ایستگاه تحقیقاتی کترا و تعیین فاصله نسبی آنها از 18 رقم تجاری، سه نوع نشانگر (مورفولوژی، ISSR و PCR-RFLP) مورد استفاده قرار گرفت.مواد و روشها: مطالعه حاضر به صورت تحقیقی سه ساله و به منظور دستیابی به اطلاعات شناسنامهای 79 ژنوتیپ محلی ناشناخته و 18 رقم تجاری مرکبات (شاهد)، تعیین روابط فیلوژنتیک و فاصله ژنتیکی آنها با یکدیگر انجام گرفت. این پژوهش بر اساس بررسی مقایسهای تعداد 19 صفت رویشی و 40 صفت زایشی و تجزیه DNA کلروپلاستی مبتنی بر نشانگرهای ISSR وPCR-RFLP نمونههای برگی انجام گرفت. میانگین سه ساله صفات مورفولوژیک بر اساس استانداردهای توصیفنامهای ثبت گردید. به منظور انجام مطالعات مولکولی، استخراج DNA از نمونههای برگی هر ژنوتیپ انجام و مقدار DNA با استفاده از نانودراپ در طول موج 260 نانومتر اندازهگیری شد. تجزیه خوشهای دادههای مورفولوژیک و مولکولی بر اساس دادههای جفت نشده (UPGMA) و ضریب تشابه جاکارد صورت گرفت و اختلاف بین ژنوتیپها بر مبنای کدگذاری و رتبهبندی آنها صورت پذیرفت. یافتهها: نتایج حاصل از تجزیه خوشهای دادههای مورفولوژی و مولکولی با نرمافزارهای NTSYS-pc و POPGENE نشان داد که کلیه ژنوتیپها میتوانند بر اساس صفات ظاهری و نشانگرهای ISSR و PCR-RFLP به ترتیب در ضریب تشابههای 40%، 53% و 60% به 12، 9 و 5 خوشه اصلی طبقهبندی شوند. طبق دادههای مورفولوژی، اولین خوشه (A) به دو زیرگروه تقسیم شد که یکی از آنها شامل 3 رقم لیمو بود. در خوشه دوم (B) همه ارقام پرتقال و نارنگی و در خوشه سوم (C) بالنگ، دارابی و گریپفروت دانکن قرار داشتند. نتایج حاصل از تجزیه نشانگر ISSR نشان داد که درصد چند شکلی ژنوتیپهای بررسی شده از 92 درصد تا 53 درصد به ترتیب برای نشانگرهای N10 و N1 متغیر بود و تعداد قابل توجهی از آنها قرابت نزدیکی با پرتقال داشتهاند. نتایج این مطالعه همچنین نشان داد که کامکوات بر اساس مشخصات مولکولی و صفات ظاهری جنسی متمایز از خانواده مرکبات است و میتواند در گروهی مجزا قرار بگیرد. از سوی دیگر، پرتقالها، گریپفروتها و پوملوها همگی در یک گروه بودند و با این واقعیت که گریپفروتها دورگهایی از پرتقال و پوملو هستند انطباق دارد. به این ترتیب، درجه مشابهت ژنوتیپهای ناشناخته محلی با یکدیگر و با رقمهای شاهد تعیین شد. علاوه بر این، تمایز سه گونه C. reticulata، C. medica و C. maxima نیز به خوبی ممکن شد.نتیجهگیری:دادههای حاصل از اندازهگیری صفات مورفولوژیک و مولکولی ژنوتیپهای ناشناخته مرکبات کلکسیون کترا میتواند اطلاعات شناسنامهای هر ژنوتیپ ناشناخته محلی را مشخص و همچنین فاصله ژنتیکی و روابط فیلوژنتیک آنها را با یکدیگر و با رقمهای تجاری تعیین کند. به این ترتیب، میتوان در آینده بر اساس نتایج حاصله، گزینشی کارآمد از والدین تلاقیها را برای دستیابی به اهداف برنامههای اصلاحی و ایجاد ارقام جدید ممکن کرد.

ارزیابی تنوع فنوتیپی، ژنتیکی و مقدار نسبی DNA هسته در فستوکای پا بلند (Festuca arundinacea)
سابقه و هدف: گونههای فستوکا در ایران رشد میکنند و پلیپلوئیدی بودن نقش مهمی در تکامل این گروه دارد. گیاه Festuca arundinacea متعلق به خانواده گراسها بوده و شامل گونههای متفاوتی است که بعنوان علوفه، چمن و همچنین برای محافظت از خاک استفاده میشوند. هدف از این تحقیق مطالعه تغییر در مقدار نسبی DNA هسته و صفات فنولوژیکی–مورفولوژیکی در ژنوتیپ-های مختلف F.arundinacea جمعآوری شده از مناطق متفاوت میباشد.مواد و روشها: این آزمایش در قالب طرح بلوک کامل تصادفی با 3 تکرار انجام شد. پارامترهای ژنتیکی برای همه صفات شامل زمان ظهور خوشه، زمان گلدهی، ارتفاع گیاه، سرعت جوانهزنی، قدرت جوانهزنی، پایداری، عملکرد علوفه، تعداد شاخه در بوته، طول سنبله، وزن هزاردانه، درصد جوانهزنی، وزن بذر در بوته، شاخص برداشت، تعداد بذر در بوته محاسبه گردید. برای اندازگیری مقدار نسبی DNA از گیاهچههای با طول عمر سه هفته، فلوروکروم DAPI و گیاه جو (Hordeum vulgare) رقم سلطان بعنوان استاندارد داخلی استفاده شد.یافتهها: ارزیابی پارامترهای ژنتیکی نشان داد تفاوت بین ضریب تغییرات ژنتیکی و فنوتیپی برای تعداد روز تا خوشهدهی، تاریخ گلدهی، تعداد ساقه در بوته، سرعت و قدرت جوانهزنی، قوه نامیه، تعداد بذر در بوته، وزن هزار دانه و ارتفاع گیاه ناچیز بوده که می-توان گفت نقش واریانس ژنتیکی بیشتر از واریانس محیطی است. از طرف دیگر مقدار وراثتپذیری صفات بین 95-66% بود. وراثت-پذیری بالا برای همه صفات بجزء طول خوشه مشاهده شد که نشان میدهد روشهای مبتنی بر گزینش برای این صفات از کارایی بالایی برخوردار است. به علت تفاوت ناچیز PCV و GCV، وراثتپذیری بالا به همراه پیشرفت ژنتیکی بالا برای صفات تعداد روز تا خوشهدهی، تاریخ گلدهی و قدرت جوانهزنی میتوان نتیجه گرفت این صفات توسط ژنهای افزایشی کنترل میشوند و میتوان آنها را در برنامههای اصلاحی از طریق گزینش بهبود داد. مشخص شد که تفاوت معنیداری بین ژنوتیپهای مورد مطالعه برای مقدار نسبی DNA هسته وجود دارد که نشاندهنده تنوع درون گونهای بالا بین ژنوتیپهای متفاوت از مناطق مختلف است. نتایج مقایسه میانگین نشان داد ژنوتیپها در 7 گروه قرار میگیرند که بالاترین و کمترین مقدار نسبی DNA هسته به ترتیب در ژنوتیپهای G13, G20, G21 وG22, G16 بود. با توجه به مطالعات گذشته، تغییر در مقدار نسبی DNA هسته را در این ژنوتیپها را میتوان به وجود کروموزوم B و تغییر در طول کروموزوم نسبت داد. نتیجهگیری: پیشنهاد میشود از صفات زمان ظهور سنبله، زمان گلدهی و قدرت جوانهزنی برای برنامههای اصلاحی استفاده شود. همچنین تغییر در مقدار نسبی DNA و صفات مورفولوژیکی-فنولوژیکی میتوانند فاکتورهای مهمی در تکامل و سازگاری این گونه به شرایط محیطی مختلف باشند.

برهمکنش آهن و سیلیسیم بر برخی از صفات بیوشیمیایی نخودفرنگی (Pisum sativum cv. Wando) در شرایط گلخانهایی
ه برهمکنش آهن و سیلیسیم بر برخی از صفات بیوشیمیایی نخودفرنگی (Pisum sativum cv. Wando) در شرایط گلخانهایی چکیدهسابقه و هدف: نخودفرنگی گیاهی علفی، یکساله و دارای طول عمر کوتاه و با نام علمی L.) Pisum sativm) که بهترین نتیجه را در آب و هوای خنک میدهد. هر چند وجود برخی فلزات سنگین همچون آهن، در محلول غذایی یا خاک برای رشد طبیعی گیاهان ضروری می-باشد غلظتهای زیاد این عناصر از طریق افزایش رادیکالهای آزاد سمی و القا تنش اکسیداتیو میتواند عاملی برای بازدارندگی رشد و ایجاد علایم سمیّت گردد. از طرف دیگر سیلیسیم میتواند باعث افزایش تولید و کیفیت محصولات در شرایط تنش ناشی از سمیّت فلزات سنگین از قبیل آهن، منگنز و آلومینیوم در گیاهان شده و بر تحریک تولید برخی آنزیمهای آنتیاکسیدانی و فتوسنتز تاثیر مثبت می گذارد. در این پژوهش اثرات سیلیسیم در حضور مقادیر مختلف آهن بر فعالیت برخی خصوضیات بیوشیمیایی در نخودفرنگی مورد بررسی قرار گرفت. مواد و روشها: آزمایش به صورت فاکتوریل و در قالب طرح بلوکهای کاملاً تصادفی با کاربرد سیلیسیم در سه سطح (0، 14 و 28 میلیگرم بر لیتر) از منبع متاسیلیکات سدیم (Na2SiO3.5H2O) و آهن در سه سطح (05/0، 1/0 و 3/0 گرم بر لیتر) از منبع کلات آهن با سه تکرار در شرایط گلخانهایی اجرا گردید. محلول غذایی مورد استفاده، محلول هوگلند تغییر یافته بود که، مقادیر یاد شده سیلیسیم و آهن به آن اضافه شده و سپس مصرف گردید. صفات بیوشیمیایی همچون کلروفیل کل، پروتئین محلول کل، میزان مالوندیآلدهید (MDA)، غلظت پراکسیدهیدروژن و فعالیت ویژه آنزیمهای کاتالاز (CAT)، گایاکول پراکسیداز (GPX) و آسکوربات پراکسیداز (APX) مورد ارزیابی قرار گرفتند. یافتهها: نتایج نشان داد که اثرات برهمکنش بین آهن و سیلیسیم بر کلروفیل کل، پروتئین، پراکسید هیدروژن، CAT و APX معنیدار شده است. با افزایش غلظت آهن میزان کلروفیل و پروتئین کاهش پیدا کرده اما سیلیسیم باعث افزایش آنها شده است و به عبارت دیگر با کاربرد سلیسیم اثرات تنش ایجاد شده توسط غلظت بالای آهن تقلیل پیدا کرد. در برهمکنش آهن و سیلیسیم بر میزان پراکسید هیدروژن، آهن باعث افزایش و سیلیسیم باعث کاهش آن در تمامی سطوح تیمارها شده است. هر دو تیمار آهن و سیلیسیم باعث افزایش فعالیت آنزیمهای CAT و APX شده است. از طرف دیگر اثرات متقابل تیمارها بر روی صفات فعالیت ویژه GPX و مالوندیآلدهید معنی دار نبود بلکه اثرات ساده آهن و سیلیسیم باعث تغییرات معنیدار در آنها شدند به طوری که آهن در هر دو باعث افزایش شده ولی سیلیسیم باعث کاهش مالوندیآلدهید و افزایش GPX شده است.نتیجهگیری: بر اساس نتایج مشاهده شده می توان گفت که غلظت های بالای آهن در گیاه نخود فرنگی، همچون اکثر گیاهان باعث افزایش تنش اکسیداتیو شده و سیلیسییم تا حدودی اثرات مخرب این نوع تنش را کاهش داده است. پس می توان از سیلسیم به عنوان یک عنصر مفید در کاهش تنشهای زیستی و غیرزیستی مورد مطالعه و استفاده قرار بگیرد. بر اساس این پژوهش میتوان گفت آهن با غلظت 1/0 گرم بر لیتر و سلیسیم با غلظت 28 میلیگرم بر لیتر بهترین اثر را در بهبود برخی ویژگیهای بیوشیمیایی نخودفرنگی داشته است.

اعتبار سنجی برخی نشانگرهای مولکولی در تعیین جنسیت ژنوتیپهای مختلف کیوی فروت حاصل گرده افشانی آزاد
سابقه و هدف: کیویفروت یکی از مهمترین محصول باغی صادراتی ایران است. اما تمایل مشتریان بین المللی به خرید میوههای کیوی متفاوت از نظر رنگ گوشت و مزه افزایش یافته است، جهت ارتقای جایگاه کیوی ایران تولید ارقام جدید اهمیت دارد. یکی از موانع اصلاح نونهالی نسبتا طولانی و همچنین دوپایگی میباشد که تا قبل از گلدهی تفاوتی ظاهری دانهال نر و ماده وجود ندارد. بنابراین، تعیین جنسیت دانهالها و جداسازی افراد نر و ماده در کوتاهترین زمان اهمیت زیادی در برنامههای بهنژادی دارد. در این پژوهش اعتبارسنجی برخی از نشانگرهای مولکولی مرتبط با جنسیت جهت تفکیک دانهالهای نر و ماده کیویفروت حاصل از گرده افشانی آزاد مورد آزمون قرار گرفت. مواد و روشها: در این پژوهش از دو نوع ژنوتیپ کیویفروت گوشت زرد (به اسامی نواب و خورشید) و یک نوع ژنوتیپ گوشت قرمز(خونی) که همگی متعلق به گونه Actinidia chinesis و دیپلوئید بودند، استفاده شد. استخراج DNA ژنومی از برگهای جوان 10 گیاه بالغ نر و ماده بالغ کیویفروت به روش CTAB با کمی تغییرات صورت گرفت. برای تعیین دقیق غلظت DNA هر نمونه، از دستگاه اسپکتروفوتومتری (Biophotometra, Plus Eppendorff) استفاده میشود و غلظتDNA در طول موج 260 نانومتر ارزیابی شد. واکنش زنجیرهای پلیمراز (PCR) در حجم 10 میکرولیتر با استفاده از دستگاه برای هر نمونه انجام گرفت. برای هر پرایمر برنامه (PCR)، غلظت ژل آگارز و مدت زمان بارگذاری بهینه استفاده شد. در نهایت، اعتبار سنجی برخی از نشانگرهای مولکولی مرتبط با جنسیت شامل سه نشانگر SCAR (SmY, SmX و SmY1) و سه نشانگر SSR (A00I, A00II, A00II) در DNA استخراج شده از برگ ژنوتیپهای مورد آزمایش، مورد بررسی قرار گرفت. یافتهها: نتایج این پژوهش نشان داد از بین چندین نشانگر SCAR و SSR که قبلا گزارش شده بود، برخی قادر به جداسازی دقیق دانهالهای نر و ماده کیویفروت نبودند. از بین سه نشانگر SCAR (SmY, SmX و SmY1) که مورد آزمون قرار گرفتند، تنها دو نشانگر SmY1 (770 bp) در افراد نر(بدون ظهور هیچ باندی در افراد ماده) و در افراد ماده و SmX (950bp) در افراد ماده (بدون ظهور هیچ باندی در افراد نر) به خوبی قادر به جداسازی دانهالهای کیویفروت شدند. با توجه به نوارهای چندشکل بدست آمده از سه نشانگر SSR (A00I, A00II, A00II) معرفی شده برای شناسایی افراد نر و ماده، فقط نشانگر A00II با باند ویژه (230bp) در افراد نر و باند ویژه (219bp) در افراد ماده قادر به تفکیک جنسیت دانهالهای کیوی فروت بود. دو نشانگر A00I و A00III در افراد نر و ماده الگوی باندی مشابه ظاهر نموده قادر به جدا سازی افراد از نظر جنسیت نبودند.نتیجه گیری: در مجموع، نتایج این پژوهش نشان داد که استفاده توام از نشانگرهای SmX, SmY1 و A00II با کمترین خطا قادر به تفکیک دانهالهای نر و ماده کیویفروت در مراحل اولیه نونهالی میباشند.

ارزیابی تاثیر تغذیه آلی، زیستی و شیمیایی بر رشد و عملکرد کرچک (Ricinus Communis)
سابقه و هدف: مصرف نهادههای شیمیایی علاوه بر آلودگی آب و خاک و کاهش کیفیت محصولات زراعی و دارویی، سبب مشکلات زیستمحیطی نیز گردیدها ست. در کشاورزی پایدار، کودهای آلی و زیستی به عنوان جایگزینی مناسب برای کودهای شیمیایی به شمار میآیند و میتوانند باعث بهبود عملکرد کمی و کیفی گیاهان شوند. با توجه به اهمیت گیاه دارویی کرچک و مصارف گسترده آن در صنایع مختلف، این تحقیق جهت دستیابی به رشد و عملکرد قابل قبول با کاربرد تلفیقی کود زیستی، ورمی کمپوست و نیتروژن اجرا شد. مواد و روشها: آزمایش به صورت فاکتوریل در قالب طرح پایه بلوکهای کامل تصادفی در سه تکرار در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه شهید مدنی آذربایجان در سال 1396 اجرا گردید. فاکتورهای مورد بررسی شامل کود نیتروژن در سه سطح، عدم مصرف ، 75 و 150 کیلوگرم در هکتار، ورمیکمپوست در سه سطح عدم کاربرد، 5 و 10 تن در هکتار، کود زیستی قارچ Piriformospora indica در دو سطح عدم تلقیح و تلقیح بودند. صفاتی مانند وزنتر و خشک گیاه، ارتفاع گیاه، تعداد برگ، سطح برگ، طول و قطر گلآذین، رنگدانههای فتوسنتزی، تعداد دانه در کپسول، تعداد کپسول در بوته، وزن هزار دانه، درصد روغن، عملکرد بذر و روغن مورد مطالعه قرار گرفتند.یافتهها: با توجه به نتایج حاصل، ترکیب تیماری عدم تلقیح با کود زیستی+ کاربرد 10 تن در هکتار ورمیکمپوست + 150 کیلوگرم در هکتار نیتروژن یشترین ارتفاع بوته، تعداد شاخه فرعی، قطر ساقه، وزنتر و خشک گیاه را در مقایسه با سایر ترکیبهای تیماری نشان داد. همچینین بیشترین میزان عملکرد بذر در ترکیب تیماری عدم تلقیح با کود زیستی+ عدم کاربرد ورمیکمپوست + 150 کیلوگرم در هکتار نیتروژن به میزان 2824 کیلوگرم و روغن در تیمار تلقیح با کود زیستی+ کاربرد 10 تن در هکتار ورمیکمپوست + 150 کیلوگرم در هکتار نیتروژن به میزان1/1185 کیلوگرم در هکتار بدست آمد. بالاترین میزان درصد روغن به میزان 30/43 درصد در ترکیب تیماری عدم تلقیح با کود زیستی+ کاربرد 10 تن در هکتار ورمیکمپوست + 150 کیلوگرم در هکتار نیتروژن و کمترین در تیمار شاهد به میزان 36 درصد حاصل شد. عملکرد روغن همبستگی مثبت و معنیداری با تعداد دانه در بوته (r=0.74) نشان داد. بالاترین ضریب تشخیص رگرسیون گام به گام (0.68) برای صفت تعداد دانه در یک بوته حاصل گردید.نتیجه گیری: صفت تعداد دانه در بوته و وزن هزاردانه و ترکیب تیماری ترکیب تیماری تلقیح با کود زیستی+کاربرد 10 تن در هکتار ورمیکمپوست+عدم کاربرد نیتروژن میتوانند انتخاب مناسبی جهت رسیدن به بالاترین عملکرد روغن گیاه کرچک در راستای کشاورزی پایدار و کاهش مصرف نیتروژن از منابع شیمیایی باشند.

سابقه و هدف: آنتوریوم با طول دوره گلدهی طولانی گیاهی متعلق به خانواده شیپوری است. تأثیر پراکسیدهیدروژن روی عمر گلجایی و کیفیت پس از برداشت گل شاخه بریده سوسن رقم مانیسا مورد بررسی قرار گرفته و نتایج نشان داده است که غلظت پائین این ماده سبب افزایش عمر گلجایی از 8/9 روز در گلهای شاهد به 8/12 روز در گلهای تحت تیمار میشود. کاربرد بنزیل آدنین نیز سبب به تأخیر انداختن پیری در رز، زنبق، لاله و جلوگیری از زرد شدن برگهای شببو و گلایول میشود. هدف از انجام این پژوهش بررسی تأثیر پراکسیدهیدروژن و بنزیلآدنین در افزایش عمر پس از برداشت آنتوریوم رقم آندرهآنوم وتعیین بهترین غلظت میباشد.مواد و روشها: به منظور بررسی اثرات غلظتهای مختلف بنزیلآدنین (از 1 تا 1000 پیپیام) و پراکسید هیدروژن (از 5 تا 100 ماکرومولار) بر خواص فیزیکوشیمیایی گل شاخه بریده آنتوریوم سه آزمایش جداگانه در قالب طرح کاملاً تصادفی با 5 تکرار اجرا گردید. شاخصهایی از قبیل عمر گلجایی، وزنتر نسبی، میزان محلول جذب شده، قند محلول و نشت یونی مورد بررسی قرار گرفت.یافتهها: نتایج حاصل از هر سه آزمایش نشان داد که غلظت 100 پیپیام بنزیلآدنین، بهطور معنیداری سبب افزایش عمر گلجایی (از 15 روز در گلهای شاهد به 6/19 روز در گلهای تیمار شده با 100 پیپیام بنزیلآدنین) شد و گلهای تیمار شده از طول عمر بالاتری نسبت به شاهد برخوردار بودند. تیمار پراکسید هیدروژن در آزمایش اول با غلظت 25 مایکرومولار اثر معنیداری بر عمر گلجایی، محلول جذب شده و وزنتر نسبی داشت در حالیکه غلظتهای بالاتر (50 و 100مایکرومولار) اثر معنیداری نداشتند.در آزمایش دوم غلظتهای 5، 15 و 25 مایکرومولار و در آزمایش سوم نیز همه غلظتهای مورد استفاده تأثیر معنیداری بر صفات مورد ارزیابی داشت. همچنین اثر همه تیمارها در آزمایش سوم بر میزان قند محلول و نشت یونی در سطح احتمال 1 درصد معنیدار بود. تأثیر غلظتهای مختلف پراکسید هیدروژن بر عمر گلجایی در هر سه آزمایش بهتر از شاهد بود.نتیجهگیری کلی: باتوجه به اثرات مثبت بنزیل آدنین و پراکسیدهیدروژن بر جذب آب، کاربرد این مواد سبب جلوگیری از کاهش آب گلهای شاخه بریده آنتوریوم میشود و این دو عامل نقش زیادی در شادابی و طول عمر گل دارند. از طرف دیگر، کاربرد این ترکیبات سبب افزایش کربوهیدرات محلول و کاهش نشت یونی در گل آنتوریوم شاخه بریده میشود. بر اساس نتایج بدست آمده، کاربرد بنزیلآدنین در غلظت 100 پیپیام و پراکسیدهیدروژن در سطوح بهینه (5، 10، 15 و 25 مایکرومولار) به افزایش شادابی و طول عمر گلهای آنتوریوم شاخه بریده کمک زیادی مینماید.

ارزیابی برخی خصوصیات فیزیولوژیک و مورفولوژیک سه ژنوتیپ انار زینتی (Punica granatum L.) تحت تنش شوری
سابقه و هدف: انار (Punica granatum L.) از خانواده Punicaceae، یک میوه محبوب گرمسیری و نیمهگرمسیری است که بومی مناطقی از ایران تا هیمالیا در شمال هندوستان میباشد. انار بهصورت گسترده در مناطق خشک و نیمهخشک ایران و همچنین مناطقی که تحت تأثیر شوری هستند، کشت و کار میشود. هدف از انجام این پژوهش، ارزیابی تحمل به شوری سه ژنوتیپ انار زینتی و معرفی متحملترین ژنوتیپ جهت توسعه در فضای سبز بود. مواد و روشها: یک آزمایش گلدانی در طول دوره نه ماهه جهت ارزیابی و مقایسه تحمل به شوری سه ژنوتیپ انار زینتی طی سال-های 97-1396 صورت گرفت. آزمایش بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با دو فاکتور 1- شوری آب آبیاری در پنج سطح 1، 3، 5، 7 و 9 دسیزیمنس بر متر و 2- ژنوتیپهای زینتی انار در سه سطح ("گلنار سروستان"، "گلنار شهداد" و "گلنار ساوه") و با چهار تکرار و مجموع 60 گلدان انجام شد. در مدت آزمایش برخی از صفات رویشی مانند درصد برگهای ریزش یافته، درصد برگهای نکروزه و سبز اندازهگیری شدند. همچنین در پایان آزمایش نیز عملکرد رویشی، وزن تر و خشک برگها و اندام هوایی، نشت یونی، محتوای نسبی آب برگ و شاخص کلروفیل و محتوی سدیم، پتاسیم، کلر و نسبت سدیم به پتاسیم در برگها اندازهگیری شدند.یافتهها: نتایج نشان داد که نوع ژنوتیپ و سطح شوری بر تغییرات صفات مورفولوژیک، فیزیولوژیک و غلظت عناصر غذایی دارای اثر معنیداری بود. در تمامی ژنوتیپهای مطالعه شده با افزایش سطح شوری، شاخصهای رشد شامل ارتفاع شاخه، قطر شاخه، تعداد برگ کل، درصد برگهای سبز، وزن تر و خشک اندام هوایی، محتوی رطوبت نسبی، شاخص کلروفیل برگها کاهش و درصد برگ-های نکروزه، درصد برگهای ریزش یافته و درصد نشت یونی افزایش یافتند. این در حالی بود که میزان کاهش و افزایش در صفات اندازهگیری شده در بین ژنوتیپهای مطالعه شده با یکدیگر اختلاف معنیداری داشت. ارزیابی غلظت عناصر غذایی در برگ نشان داد که در تمامی ژنوتیپهای مطالعه شده، بیشترین مقدار کلر و سدیم، نسبت سدیم به پتاسیم در شوری 9 دسیزیمنس بر متر، مشاهده شد. همچنین، در تمام ژنوتیپهای مورد بررسی کاهش ارتفاع و سطح برگ بهترتیب از شوری 5 و 7 دسیزیمنس بر متر بهطور معنیدار شروع به کاهش کرد. افزایش نکروزگی، کاهش درصد برگ سبز و کاهش محتوای نسبی آب برگ در ژنوتیپ "گلنار سروستان" از شوری 5 دسیزیمنس بر متر و در ژنوتیپهای "گلنار شهداد" و "گلنار ساوه" از شوری 7 دسیزیمنس بر متر مشاهده شد. در ژنوتیپهای "گلنار سروستان" و "گلنار ساوه" در شوری 7 دسیزیمنس بر متر و در ژنوتیپ "گلنار شهداد از شوری 9 دسیزیمنس بر متر ریزش برگها افزایش یافت. در دو ژنوتیپ "گلنار سروستان" و "گلنار شهداد" نشت یونی در شوری 7 دسیزیمنس بر متر و در ژنوتیپ "گلنار ساوه" در شوری 5 دسیزیمنس بر متر بهطور معنیداری نسبت به شوریهای سطوح پائینتر (1و 3 دسیزیمنس بر متر) افزایش یافت. در سطوح بالای شوری (7 و 9 دسیزیمنس بر متر) ژنوتیپهای "گلنار شهداد" و "گلنار ساوه" نسبت به "گلنار سروستان" بهطور معنیداری جذب سدیم، کلر و نسبت سدیم به پتاسیم کمتری را نشان دادند. نتیجهگیری: با توجه به نتایج بررسی حاضر در مجموع ژنوتیپهای "گلنار شهداد" و "گلنار ساوه" بهتریتیب به عنوان متحملترین و حساسترین ژنوتیپ نسبت به تنش شوری انتخاب شدند. ژنوتیپ "گلنار شهداد" توانست از طریق افزایش جذب پتاسیم در مقابل سدیم و در نتیجه حفظ خصوصیات رشدی خود ، به خوبی شوری تا 7 دسیزیمنس بر متر را تحمل نماید.

تاثیر اسید سالیسیلیک بر کارایی فیزیولوژیکی نیتروژن و مصرف آب جو در شرایط قطع آبیاری
سابقه و هدف: تنش بر فعالیتهای فیزیولوژیکی گیاه تاثیر گذاشته و مانع رشد و تقسیم سلولی میشود و در نهایت سبب کاهش عملکرد در گیاهان میشود. کاربرد موادی که بتواند اثرات خشکی را کم کند میتواند در جلوگیری از کاهش بیش از حد عملکرد گیاهان زراعی موثر باشد. هدف از این مطالعه تاثیر سالسیلیک اسید در کاهش اثرات قطع آبیاری بر عملکرد، کارآیی فیزیولوژیکی نیتروژن، کارایی مصرف آب و برخی خصوصیات فیزیولوژیکی جو بود.مواد و روشها: به منظور بررسی تاثیر سالیسیلیک اسید بر کارآیی فیزیولوژیکی نیتروژن و مصرف آب و برخی خصوصیات جو در شرایط تنش خشکی، آزمایشی در سال زراعی 1396 به صورت اسپلیت پلات در قالب طرح پایه بلوکهای کامل تصادفی با سه تکرار در شهرستان سبزوار انجام شد. تیمارها شامل آبیاری در دو سطح تنش خشکی (قطع آبیاری از مرحله آبستنی(کد 49 زادوکس) تا برداشت) و آبیاری کامل و تیمار محلول پاشی سالیسیلیک اسید در چهار سطح صفر (شاهد)، دو، چهار و شش میلی مولار در مرحله ساقهدهی(کد 39 زادوکس) روی جو اجرا شد. یافتهها: نتایج نشان داد که در هر دو شرایط آبیاری کامل و تنش خشکی بیشترین تعداد سنبله، تعداد دانه، عملکرد دانه و عملکرد بیولوژیکی در متر مربع به ترتیب در تیمار محلول پاشی دو تا شش میلی مولار سالسیلیک اسید و کمترین آن مربوط به تیمار شاهد بود. کمترین کارایی مصرف آب در شرایط آبیاری کامل و بدون مصرف سالیسیلیک اسید حاصل شد و با مصرف سالیسیلیک اسید تا شش میلی مولار کارایی مصرف آب افزایش یافت. در هر دو شرایط آبیاری کامل و قطع آبیاری، بیشترین و کمترین عملکرد دانه به ترتیب از تیمار محلول پاشی شش میلی مولار سالیسیلیک اسید و تیمار شاهد (بدون محلول پاشی سالیسیلیک اسید) بدست آمد. در شرایط آبیاری کامل و تنش خشکی، تیمار محلول پاشی شش میلی مولار سالسیلیک اسید در مقایسه با شاهد (بدون محلول پاشی سالیسیلیک اسید)، عملکرد دانه را به ترتیب به میزان 44 و 48 درصد افزایش داد. نتیجهگیری: به نظر میرسد تاثیر محلول پاشی سالیسیلیک اسید در شرایط تنش خشکی در مقایسه با تیمار قطع آبیاری، در بهبود کارآیی مصرف آب موثرتر از شرایط آبیاری کامل بوده استکارایی فیزیولوژیکی نیتروژن با افزایش مصرف سالیسیلیک اسید افزایش یافت و مصرف شش میلیگرم سالسیلیک اسید با نسبتهای 6/124 و 1/132 به ترتیب در هر دو شرایط آبیاری و قطع آبیاری بیشترین مقدار کارایی فیزیولوژیکی نیتروژن را داشت. شرایط قطع آبیاری باعث شده تا انتقال مجدد نیتروژن افزایش یابد و همچنین کارایی فیزیولوژیکی نیتروژن نیز به 95/113 افزایش یافت. از طرفی، اسید سالیسیلیک موجب تعدیل تنش خشکی از طریق افزایش کارایی فیزیولوژیکی و انتقال مجدد نیتروژن باعث بهبود عملکرد دانه گندم در شرایط تنش در مقایسه با آبیاری کامل گردید.

بررسی تأثیر تنش کم آبی بر رشد میزان اسانس و برخی صفات فیزیولوژیک چهار رقم ریحان (Ocimum basilicum L.)
سابقه و هدف: یکی از مهمترین عوامل محیطی که پراکنش، رشد و تولید موفق محصولات گیاهی را تحت تأثیر قرار میدهد، میزان آب در دسترس گیاه است. خشکی بهعنوان مهمترین عامل غیرزنده محدودکنندهی رشد و عملکرد گیاهان، باعث بروز یا تشدید سایر تنشها مخصوصاً تنش کمبود عناصر غذایی در گیاه نیز میشود. از طرفی دیگر تحت شرایط تنش کم آبی، گیاهان مقادیر مشابهی از متابولیتهای اولیه در مقایسه با شرایط بدون تنش تولید میکنند و چون تحت شرایط تنش، گیاه میزان تولید بیوماس و ماده خشک خود را کاهش میدهد، لذا مقادیر زیادی از این متابولیتهای اولیه به سمت تولید متابولیتهای ثانویه میروند. پژوهش حاضر بهمنظور ارزیابی میزان رشد، اسانس و برخی صفات فیزیولوژیکی چهار رقم ریحان تحت شرایط تنش کم آبی انجام گرفت. مواد و روشها: آزمایش بهصورت فاکتوریل در قالب طرح پایهی کاملا ً تصادفی در شرایط گلخانه در سال 1396 انجام گرفت. چهار رقم تجاری ریحان (شامل ارقام سبز ایرانی، بنفش ایرانی، گنوس و آمیتیست) بهعنوان فاکتور اول و تنش خشکی در چهار سطح (شامل آبیاری درحد 100، 85، 70 و 55 درصد ظرفیت زراعی) بهعنوان فاکتور دوم در نظر گرفته شد. ویژگیهایی مانند وزن تر و خشک گیاه، وزن تر و خشک ریشه، درصد اسانس، عملکرد اسانس، کلروفیل، مالوندیآلدئید، پرولین، کاتالاز، پراکسیداز، آسکوربات پراکسیداز، فتوسنتز، هدایت روزنهای و مقاومت روزنهای اندازهگیری شد. دادهﻫﺎی بهدست آمده از آزمایش ﺑﺎ اﺳﺘﻔﺎده از ﻧﺮم افزار Minitab ﻣﻮرد ﺗﺠﺰﻳهی آﻣﺎری ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﻣﻘﺎﻳﺴهی ﻣﻴﺎﻧﮕﻴﻦﻫﺎ ﻧﻴﺰ ﺑﺎ اﺳﺘﻔﺎده از روش آزمونLSD در سطح 05/0 انجام ﮔﺮﻓﺖ.یافتهها: نتایج نشان داد تنش کم آبی باعث کاهش وزن تر و خشک اندام هوایی و فتوسنتز در همه ارقام شد. علاوه بر این، تحت تأثیر تنش خشکی، وزن تر و خشک ریشه و هدایت روزنهای کاهش و میزان پرولین، مالوندیآلدئید، کلروفیل و آنزیمهای آنتیاکسیدان افزایش نشان دادند. با این وجود شدت تغییرات این صفات در ارقام مورد مطالعه متفاوت بود و در اغلب موارد شدت کاهش صفات مختلف در رقم گنوس کمتر از سایر ارقام بود. در مقایسه بین ارقام نیز، ارقام سبز ایرانی و گنوس بیشترین میزان وزن تر و خشک بوته را دارا بودند. علاوه بر این، بیشترین درصد اسانس مربوط به رقم گنوس بود.نتیجه گیری: رقم گنوس بیشترین وزن تر و خشک اندام هوایی، بالاترین درصد اسانس و بیشترین میزان هدایت روزنهای را به خود اختصاص داد و بهعنوان رقم متحمل جهت کشت در شرایط کم آبی بهمنظور استحصال اسانس توصیه میشود.واژههای کلیدی : پارامترهای فیزیولوژیک، تنش کم آبی، اسانس، پارامترهای فتوسنتزی

تاثیر بنزیل آدنین بر کربوهیدراتهای غیر ساختاری و محلول در آب در سبز فرش فستوکای بلند تحت تنش خشکی
سابقه و هدف: کاربرد سایتوکنین خارجی بر روی برگ سبزفرشها و اثر آن در تعدیل تنش خشکی به شکل خیلی محدود گزارش شده است که با بهبود مقاومت به خشکی همراه بوده است. اما اثر بنزیلآدنین در محتوای کل کربوهیدراتهای غیر ساختاری و کربوهیدراتهای محلول در آب برگ سبز فرش فستوکایبلند (Festuca arundinacea Schreb. ) تحت شدتهای مختلف تنش خشکی مورد بررسی قرار نگرفته است که از جمله اهداف ما در این آزمایش است.مواد و روشها: یک دوره تنش یک ماهه بر روی گونه فستوکایبلند که به صورت همگروهی تکثیر و طی یکسال در گلخانه مستقر گردید به صورت طرح کامل تصادفی با حداقل سه تکرار مورد بررسی قرار گرفت که شامل تیمارهای آبیاری در چهار سطح (100%، 75%،50% و 25% ظرفیت مزرعه) بود و برروی تیمارهای ذکر شده محلول پاشی برگی 50 میکرومولار بنزیلآدنین با فاصله هر 7 روز یکبار انجام شد. در نمونه های شاهد تحت تنش خشکی، برای مقایسه، محلول پاشی با بنزیل آدنین صورت نگرفت.یافته ها: تیمار با بنزیلآدنین تنش شدید 25% ظرفیت مزرعه را به شکل جالبی تعدیل نمود و منجربه بازگشت مشهود برگها به حالت عادی رشد شد. بررسی کربوهیدراتهای محلول در آب (WSCs) نشان از افزایش چندین برابری نسبت به شاهد دارد که این افزایش محتوای کربوهیدراتهای محلول در آب در تنش 50% و 25% ظرفیت مزرعه نسبت به دیگر تیمارها مشهودتر است. محلولپاشی با بنزیلآدنین اگرچه که این افزایش تولید قندهای محلول را به سطح مشاهده شده در شاهد نرساند اما محتوای کربوهیدراتهای محلول در آب را به نصف مقدار تولید شده نسبت به تنش خشکی 50% و 25% ظرفیت مزرعه بدون محلولپاشی کاهش داد. تنش خشکی باعث کاهش میزان کلروفیل و نشاسته ذخیرهای برگ شد. این کاهش کلروفیل در تیمار 25% ظرفیت مزرعه، به شکل معنی داری در گیاهان تحت تنش که با 50 میکرومولار بنزیل آدنین تیمار شده بودند جبران شد. فعالیت آنزیمهای آلفا و بتا آمیلاز برگ با شروع تنش خشکی و محلولپاشی بنزیلآدنین در مقایسه با شاهد به صورت معنیداری افزایش و سپس کاهش یافت.نتیجه گیری: کاهش شدید محتوای آب خاک و سپس آب گیاه منجر به فعال شدن آنزیمهای تجزیه کننده نشاسته و افزایش در کربوهیدرات های محلول در آب میشود که هم به حفظ هموستازی سلول برگ کمک کرده و هم با فراهم نمودن ATP به تحمل بیشتر گیاه به تنش خشکی کمک خواهد نمود. قسمتی از این تجزیه نشاسته به وسیله افزایش فعالیت آنزیمهای آلفا و بتا آمیلاز در برگ قابل توجیه خواهد بود. کاربرد 50 میکرومولار بنزیلآدنین به شکل معنیداری از شدت تنش وارد شده به گیاه کاست. این تیمار باعث احیاء کلروفیل برگ و بهبود فتوسنتز در تنش 25% ظرفیت زراعی شد و از تجزیه بیشتر نشاسته و تبدیل شدن آن به فروکتوز، گلوکز و ساکارز کاست تا بدین صورت گیاه منابع کربوهیدراتی خود را حفظ نماید.کلید واژهها: سایتوکنین، فستوکای بلند، کربوهیدرات های محلول در آب، مقاومت به خشکی.

مطالعه توارث مقاومت به زنگ قهوهای در برخی ژنوتیپهای گندم نان به روش دایآلل
سابقه و هدف: گندم در طول دورهی رشد مورد حمله بسیاری از عوامل بیماریزا از جمله زنگها قرار میگیرد. عامل بیماری زنگ قهوهای گندم قارچی با نام Puccinia recondita f. sp. Tritici میباشد که به نام زنگ برگی نیز معروف است. مهمترین راه کنترل بیماری زنگ قهوهای استفاده از ارقام مقاوم اصلاح شده است که بهطور موثری باعث کنترل این بیماری شده و خسارت آن را کاهش میدهد. بررسی نحوهی توارث مقاومت به زنگ قهوهای در ارقام مختلف گندم و برآورد پارامترهای ژنتیکی با استفاده از تلاقی دایآلل از اهداف این پژوهش میباشد. مواد و روشها: شش رقم گندم نان شامل ژنوتیپهای گنبد، شیرودی، دریا، اترک و مغان3 به همراه رقم حساس بولانی و 15 نتاج F1 حاصل از تلاقی دایآلل یک طرفه مورد بررسی قرار گرفت. این پژوهش در قالب طرح بلوکهای کامل تصادفی با سه تکرار در شرایط مزرعه و گلخانه انجام شد. صفات مورد بررسی شامل تیپ آلودگی، شدت آلودگی، سطح زیر منحنی پیشرفت بیماری و دوره کمون بودند. تجزیه دادهها با استفاده از نرمافزارهای SAS ver 9.1 و Excel و برآورد پارامترهای ژنتیکی و نحوه توارث صفات مورد بررسی از طریق تجزیهی دای آلل با استفاده از روش دوم گریفینگ با مدل ثابت و روش جینکز و هیمن صورت گرفت.یافتهها: نتایج تجزیه واریانس نشان داد که بین ژنوتیپها برای تمام صفات اختلاف معنیداری وجود داشت که نشاندهنده وجود تنوع ژنتیکی برای این صفات بود و در نتیجه میتوان تجزیه ژنتیکی را انجام داد. میانگین مربعات ترکیبپذیری عمومی و خصوصی برای تمامی صفات معنیدار بود که مبین اهمیت هر دو اثرات افزایشی و غالبیت ژنها در توارث این صفات بود. نسبت بیکر، برای صفت دوره کمون بیانگر اهمیت یکسان اثرات افزایشی و غالبیت بود و برای صفات مذکور دیگر بیانگر اهمیت بیشتر اثرات افزایشی بود. برآورد پارامترهای ژنتیکی به روش جینکز و هیمن نشان داد که برای هر چهار صفت، مقدار جزء افزایشیD نسبت به اجزاء غالبیت(H1 و H2)کمتر بود که بیانگر اهمیت بیشتر جزء غالبیت نسبت به جزء افزایشی در کنترل این صفات میباشد. علامت F برای تمام صفات مثبت بود، در نتیجه میتوان گفت که فراوانی آللهای غالب نسبت به آللهای مغلوب بیشتر بود. آماره wr-vr برای تمام صفات مورد بررسی معنیدار شد که به منزله وجود اثرات غیرآللی (اپیستازی) میباشد. به عبارت دیگر معنیدار شدن این آماره نشان دهنده عدم صحت مدل افزایشی- غالبیت برای صفات میباشد.نتیجهگیری: براساس صفات ارزیابی شده در مزرعه ؛ رقم اترک و در گلخانه، رقم گنبد مقاومت خوبی به بیماری زنگ قهوهای داشتند. وراثت پذیری خصوصی صفات بین 51 تا 86 درصد برآورد گردید.برآورد پارامتر های ژنتیکی نشان داد که مدل افزایشی- غالبیت کفایت نمی کند. به عبارت دیگر حداقل دوژن مقاومت در ارقام مقاوم قابل پیش بینی است.
